摘 要:目的探究PI3K抑制剂TGX-221对人骨肉瘤细胞增殖、周期及凋亡的影响。方法培养人骨肉瘤U2OS和MG-63细胞株,分为对照组和实验组,实验组加入10μmol/L的TGX-221,对照组加入相同剂量的DMSO。处理48 h后,CCK8法检测TGX-221对人骨肉瘤细胞活力及增殖的影响;流式细胞术检测TGX-221对人骨肉瘤U2OS和MG-63细胞的细胞周期及凋亡的作用。结果CCK8检测结果显示,TGX-221对人骨肉瘤细胞有显著的增殖抑制作用,且具有时间和浓度依赖性;流式细胞术结果显示,TGX-221将U2OS和MG-63细胞周期阻滞在G1期,并诱导人骨肉瘤细胞凋亡,具有显著的抗肿瘤作用。结论PI3K抑制剂TGX-221能够抑制人骨肉瘤细胞的增殖、阻滞细胞周期并诱导细胞凋亡,可能成为一种潜在的抗肿瘤药物。Objective To investigate the effect of P13K inhibitor TGX-221 on proliferation,cycle and apoptosis of human osteosarcoma cells.Methods Human osteosarcoma U2OS and MG-63 cell lines were cultured and divided into the control group and the experimental group.The experimental group was added with 10μmol/L of TGX-221,and the control group was added with the same amount of DMSO.After 48 hours of treatment,the effect of TGX-221 on the viability and proliferation of human osteosarcoma cells were detected by CCK8 method.The effect of TGX-221 on the cycle and apoptosis of human osteosarcoma U2OS and MG-63 cells were detected by flow cytometry.Results CCK8 showed that TGX-221 significantly inhibited the proliferation of human osteosarcoma cells in a time and concentration dependent manner.Flow cytometry revealed that TGX-221 blocked U2OS and MG-63 cell cycle in G1 phase.In addition,TGX-221 also induced apoptosis in human osteosarcoma cells,demonstrating a significant anti-tumor effect.Conclusion PI3K inhibitor TGX-221 can inhibit the proliferation of human osteosarcoma cells,and it can block cell cycle and induce apoptosis,which may become a potential anti-tumor drug.
