摘 要:目的:通过检测温阳健脑方不同剂量对血管性认知障碍(VCI)模型大鼠海马组织血管内皮生长因子(VEGF)、β-内啡肽(β-EP)和内皮素-1(ET-1)的影响,观察其对VCI大鼠的脑保护作用。方法:将30只按照改良2VO法造模成功的VCI模型大鼠,按照随机数字表法随机分为模型组、吡拉西坦组、温阳健脑低剂量组、温阳健脑高剂量组,每组各6只,以及6只同时喂养的正常大鼠作正常对照组。造模成功后除正常组及模型组外,其余各组均按照实验设计时规定的用药及剂量连续灌胃14 d后,断头处死各组大鼠,ELISA法检测大鼠海马组织β-EP和VEGF的含量,放射免疫法检测ET-1的含量。结果:与正常组比较,模型组海马组织VEGF下降,并有显著性差异(P<0.01);与模型组比较,各给药组VEGF水平上升,温阳健脑高剂量组有显著性差异(P<0.01),其余两组差异不显著。各组大鼠海马组织β-EP含量虽各有不同,但无统计学差异差异(P>0.05)。与正常组比较,模型组海马组织ET-1水平上升,并有显著性差异(P<0.01);与模型组比较,各给药组较模型组ET-1水平均有下降趋势,除吡拉西坦组外均有显著性差异(P<0.05);尤其温阳健脑高剂量组显著(P<0.01)。结论:温阳健脑方对VCI模型大鼠具有脑保护作用,其机制可能通过升高大鼠海马组织VEGF和降低ET-1的含量有关,但与β-EP的含量无关。Objective:To explore the protection of different doses of Wenyang Jiannao Recipe on brain tissue of rats with vascular cognitive impairment(VCI)by detecting the effects on vascular endothelial growth factor(VEGF),β-endorphin(β-EP)and endothelin-1(ET-1)in rats hippocampus.Methods:Thirty healthy rats were made models by 2VO method were randomly divided into model group(MG),piracetam group(PG),Wenyang Jiannao low-dose group(WLG),Wenyang Jiannao high-dose group(WHG),6 rats in each group,and 6 normal rats fed simultaneously as normal control group(NG).After successful modeling,except for NG and MG,the other groups were continuously administered for 14 days according to the medication and dosage prescribed in the experimental design.The rats in each group were killed by decapitation,theβ-EP and VEGF levels of hippocampus were detected by ELISA method.,the content of ET-1 was determined by radioimmunoassay.Results:Compared with NG,VEGF in the hippocampus of MG decreased and there was a significant difference(P<0.01);compared with MG,the VEGF level in each administration group increased,and there was a significant difference in WHG(P<0.01),there was no difference between the other two groups.Althogh there was different inβ-EP content in the hippocampus of every group rats,but the differences were no significant(P>0.05).Compared with NG,the level of ET-1 in the hippocampus of MG increased and there was a significant difference(P<0.01);compared with MG,the ET-1 level of each administration group had a downward trend,except for PG,there were significant differences(P<0.05,P<0.01).Conclusion:Wenyang Jiannao Recipe has brain protective effect on VCI model rats.The mechanism may be related to increasing VEGF and reducing ET-1 content in hippocampus of rats,but it has nothing to do withβ-EP content.
