摘 要:目的观察补精益视片对SD大鼠慢性高眼压(EIOP)模型初级视皮质(PVC)的PI3K/Akt通路磷酸化FoxO1和IKK2表达的影响,并对其作用机理进行初步探讨。方法采用烙闭3支上巩膜静脉法,制作大鼠慢性EIOP模型,随机分为3组:对照组、模型组、给药组。连续灌胃8周,并于8周末处死大鼠,观察其对大鼠眼压(IOP),初级视皮质PI3K/Akt通路p-FoxOl及IKK2表达的影响。结果①造模后大鼠IOP明显升高,造模后8周(实验结束时)各组IOP仍较高,说明模型成功,说明造模成功,补精益视片有降低IOP作用(P<0.05)。②造模后8周,模型组p-FoxO1 JKK2较对照组及给药组含量高,差异有统计学意义(均为P<0.05);给药组p-FoxO1 JKK2含量高于对照组,差异有统计学意义(P<0.05)。结论补精益视片可以保护SD大鼠慢性EIOP模型视功能,表现为:降低眼压,下调p-FoxO1和IKK2的表达。Objective To observe the effect of traditional BuJingYiShi tablets on primary visual cortex(PVC)changes in SD rat model of chronic elevated intraocular pressure(EIOP),and explore the mechanism of it initially.Methods By unilaterally cauterizing 3 episcleral vessels establish the rat model of chronic EIOP rats were divided into 3 groups randomly:control group,model group and treatment group.After given BuJingYiShi tablets or normal saline for 8 weeks,the rats were put to death.To observe the effection of BuJingYiShi tablets on the EIOP SD rats model's intraocular pressure(IOP),niss body and PI3K/Akt signal pathway's p-FoxO1 and IKK2 in primary visual cortex.Results①IOP of the EIOP SD rats model's increased significantly after modeling,and still higher 8 weeks after modeling(at the end of the experiment),indicating that the model was successful,indicating that the model was successful,and BuJingYiShi tablets had the effect of reducing IOP(P<0.05).②8 weeks after-modeling,the Immunohistochemical detection of primary visual cortex showed that the control group's expression on p-FoxO1>IKK2 was the highest,compared with the treatment group and modle group,there were statistically differences(all P<0.05);The Immunohistochemical detection of primary visual cortex showed that the modle group's expression on p-FoxO1>IKK2 was the higher,compared with the control group,there were statistically differences(all P<0.05).Conclusion BuJingYiShi tablets can protect the visual function from elevating IOP>by reducing the expression of p-FoxO1 and IKK2 in PVC.
