摘 要:目的探讨前列腺癌组织中PCDH10基因甲基化状态及意义。方法选取2014年3月~2016年5月在我院行腹腔镜前列腺癌根治术或经尿道前列腺电切术患者标本121例,其中前列腺癌59例(前列腺癌组),良性前列腺增生62例(前列腺增生组),采用甲基化特异性聚合酶链式反应(polymerase chain reaction,PCR)检测甲基化情况。结果 PCDH10基因在22RV1、PC3、DU145和LNCaP四株前列腺癌细胞系中的启动子CpG岛均有不同程度的甲基化;前列腺癌组有66.10%出现PCDH10基因甲基化,明显高于前列腺增生组,差异比较有统计学意义(P<0.05);前列腺癌患者PCDH10基因甲基化与年龄、PSA和Jewett分期无关(P>0.05);Gleason评分8~10分前列腺癌患者PCDH10基因甲基化比例为76.32%,明显高于Gleason评分4~7分患者(P<0.05)。PCDH10基因甲基化患者术后的生化复发生存率显著减少(χ~2=4.116,P=0.043),总体生存率明显提高(χ~2=6.731,P=0.010)。结论 PCDH10基因甲基化可能参与了前列腺癌的发生发展,值得进一步研究。Objective To explore the methylation status of PCDH10 gene in prostate cancer and its significance. Methods A total of 121 patients with laparoscopic radical prostatectomy or transurethral resection of prostate from March 2014 to May 2016 in our hospital were selected, among them, 59 cases of prostate cancer(prostate cancer group), 62 cases of benign prostatic hyperplasia(prostatic hyperplasia group), methylation specific polymerase chain reaction(PCR) was used to detect methylation. Results PCDH10 gene in 22 RV1, PC3, DU145 and LNCaP 4 prostate cancer cell lines the promoter CpG islands had varying degrees of methylation;The methylation of PCDH10 gene in prostate cancer group was 66.10%, which was significantly higher than that in prostatic hyperplasia group, and the difference was statistically significant(P<0.05);Methylation of PCDH10 gene in prostate cancer was not associated with age, PSA, and Jewett stage(P<0.05);Gleason score of 8 to 10 prostate cancer patients with PCDH10 gene methylation ratio was 76.32%, significantly higher than the Gleason score of 4 to 7 patients(P<0.05). The survival rate of biochemical recurrence after PCDH10 methylation was significantly decreased(χ~2=4.116, P=0.043) and overall survival was significantly improved(χ~2=6.731, P=0.010). Conclusion Methylation of PCDH10 gene may be involved in the occurrence and development of prostate cancer, which is worthy of further study.
