摘　　要：头颈肿瘤是严重威胁人类健康的疾病之一,其中90%都是头颈鳞癌。头颈鳞癌的发生率和致死率都位于前列,复发和转移是导致低存活率的主要原因。研究表明肝细胞生长因子(HGF)及其受体C-Met对肿瘤细胞的形态发生、生长和运动有密切关系。本研究探讨了对头颈鳞癌侵袭转移的影响,通过体外培养头颈鳞癌细胞,采用MTS法检测了外源HGF对肿瘤细胞增殖能力的影响,细胞划痕实验和Transwell小室实验测试了外源HGF对肿瘤细胞侵袭转移能力的影响,免疫沉淀法(Western blotting)检测了在不同浓度HGF诱导下,肿瘤细胞体内C-Met表达量的变化。结果表明外源HGF对头颈鳞癌细胞的侵袭转移能力有显著的增强作用,C-Met的表达量在HGF的诱导下也有明显提升,这些结果为探究HGF/C-Met系统对头颈鳞癌的侵袭转移作用及机制提供了理论依据。Head and neck cancer is one of diseases that seriously risk human health, and 90% of head and neck cancer are Head and Neck Squamous Cell Carcinoma(HNSCC). Due to cancer reappearance and metastasis, both of the incidence of HNSCC and its death rate stand out in the front of all other diseases. Some studies show that hepatocytegrowthfactor(HGF)and its receptor C-Met played important role in the morphogenesis,growth and motion of cancer cells. By MTS cell proliferation assay, scratch wound migration assay and transwell assay, western blotting,we studied the function of HGF in the invasion and metastasis of HNSCC in vitro cells, detected its expression of C-Met with HGF stimulation. The result shows that, the invasion and metastasis of HNSCC cells have been greatly improved with HGF stimulation, the expression of C-Met has also increased with HGF stimulation. That provides evidence and foundation for illustrating the mechanism of invasion and metastasis of HNSCC.
