摘 要:目的探讨口腔鳞状细胞癌(OSCC)中转化因子2β(Tra2β)的表达及临床意义。方法利用免疫组化检测64例OSCC中Tra2β蛋白的表达,并分析Tra2β蛋白表达与临床病理参数间的关系。Western blot和实时荧光聚合酶链反应(RT-PCT)检测22例OSCC肿瘤和癌旁组织中Tra2β蛋白和mRNA的表达情况。Kaplan-Meier分析Tra2β蛋白的表达对OSCC患者生存率的影响。Cox多因素生存分析Tra2β蛋白表达对OSCC患者预后的影响。结果免疫组化检测结果表明Tra2β蛋白在OSCC中表达,并且它的阳性表达与分化程度和临床分期有关。实验结果表明,Tra2β蛋白和mRNA在OSCC中的表达趋势一致,肿瘤组织中Tra2β蛋白和mRNA的表达均高于癌旁组织。Kaplan-Meier分析表明,Tra2β蛋白高表达患者的预后较差。Cox多因素分析表明,Tra2β蛋白是OSCC患者预后评估的一个指标。结论Tra2β可能为OSCC治疗的一个新的分子靶点。Objective This study aimed to investigate the expression and clinical signifi cance of Transformer 2β(Tra2β)in oral squamous cell carcinoma(OSCC).Methods Sixty-four samples of OSCC were subjected to immunohistochemistry to analyze the expression of Tra2βprotein in OSCC and to explore the relationship between the expression of Tra2βprotein and different clinical pathological parameters.Twenty-two pairs of fresh OSCC tissues and adjacent tissues were used to explore the expression of Tra2βmRNA and protein through real-time polymerase chain reaction and Western blot analyses.The Kaplan-Meier method was used to estimate and validate the effect of Tra2βprotein on the survival rate of patients with OSCC.Cox multivariate regression was used to analyze the effect of Tra2βprotein on the prognosis of patients with OSCC.Results Immunohistochemistry results revealed that Tra2βprotein was expressed in OSCC,and the positive expression of Tra2βprotein was significantly associated with the differentiation degree and clinical stage of OSCC.Moreover,the expression trends of Tra2βprotein and mRNA were consistent,and the expression levels of Tra2βprotein and mRNA in OSCC tissues were higher than those in adjacent tissues.Kaplan-Meier survival analysis showed that patients with OSCC and Tra2βprotein expression had poor prognoses.Cox multivariate regression analysis demonstrated that the Tra2βprotein was an independent prognostic factor of OSCC.Conclusion Tra2βmay be a new molecular target for the treatment of OSCC.
