摘  要:目的探讨尼莫地平对脑出血大鼠脑组织含水量及白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平的影响。方法72只SD大鼠以随机数表法分为假手术组、脑出血模型组和尼莫地平治疗组,每组24只。采集自体股动脉取血50μL注入大鼠基底节区复制实验性脑出血模型。尼莫地平治疗组在造模后接受尼莫地平5 mg·kg-1腹腔注射治疗,每12 h注射1次,直至处死前12 h。假手术组制备过程相同,基底节区注入等体积9 g·L-1的NaCl溶液。术后72 h,采用干湿重法检测各组脑组织含水量,酶联免疫法检测各组大鼠脑组织匀浆液中IL-6、TNF-α水平。结果脑出血模型组脑组织含水量高于假手术组,差异有统计学意义(P=0.0081)。尼莫地平治疗组脑组织含水量低于脑出血模型组,差异有统计学意义(P=0.0342)。脑出血模型组脑组织匀浆液IL-6、TNF-α水平高于假手术组,差异有统计学意义(P=0.0064、0.0073)。尼莫地平治疗组脑组织匀浆液IL-6、TNF-α水平低于脑出血模型组,差异有统计学意义(P=0.0345、0.0171)。结论尼莫地平具有减轻脑出血后水肿、抑制炎症因子的作用。Objective To investigate the effects of nimodipine on brain water content,interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)in rats with intracerebral hemorrhage.Methods Seventy-two SD rats were randomly divided into sham operation group,cerebral hemorrhage model group and nimodipine treatment group,24 rats in each group.The experimental cerebral hemorrhage model was established by injecting 50μL of autologous femoral artery blood into the basal ganglia of rats.The nimodipine treatment group was given 5 mg·kg-1 intraperitoneal injection of nimodipine every 12 hours until 12 hours before experiment.The sham-operated group had the same preparation process,and the basal ganglia was injected with saline of equal volume.At 72 hours after operation,the rats were decapitated and the related indexes were detected.At 72 hours after operation,the water content of brain tissue was measured by dry-wet weight method,and the contents of IL-6 and TNF-αin brain homogenate were detected by enzyme-linked immunosorbent assay.Results The water content of brain tissue in the model group was higher than that in the sham operation group(P=0.0081),and that in the nimodipine treatment group was lower than the model group(P=0.0342).The level of IL-6,TNF-αin brain homogenate of cerebral hemorrhage model group were higher than sham operation group(P=0.0064,0.0073).IL-6,TNF-αin brain homogenate of nimodipine treatment group were lower than cerebral hemorrhage model group(P=0.0345,0.0171).Conclusion Nimodipine has a better effect on reducing edema and inhibiting inflammatory factors after cerebral hemorrhage.
