摘 要:以不同浓度铅处理的向日葵HA300自交系幼苗为材料,运用傅里叶红外光谱(FTIR)技术分析铅胁迫下向日葵幼苗蛋白质二级结构的变化。结果表明:铅胁迫后向日葵幼苗根、茎、叶及全株的蛋白二级结构在酰胺Ⅰ区都有变化。较对照而言,根、茎在600 mg/L铅胁迫下蛋白二级结构均较稳定,有序结构分别增加了15.01%、16.45%;叶的蛋白二级结构在铅胁迫后无序结构相对含量增加,蛋白二级结构稳定性下降;在全株变化中,600 mg/L铅胁迫下蛋白二级结构较稳定,说明向日葵HA300幼苗能在一定浓度的铅胁迫下维持其蛋白二级结构的稳定性。本研究结果可以为逆境下向日葵幼苗生理机制的研究提供一定的科学依据。The sunflower HA300 inbred line seedlings were treated with different concentrations of lead were used as materials.Then the changes of protein secondary structure were analyzed by Fourier transform infrared spectroscopy(FTIR).The results showed that the protein secondary structure of roots,stems,leaves and whole plants of sunflower seedlings were changed in the amideⅠregion under lead stress.Compared with the control group,the protein secondary structure of roots and stems were stable under 600 mg/L lead stress,in addition,the ordered structure was increased 15.01%and 16.45%respectively.The disordered structure of leaves was increased under lead stress,which the stability of protein secondary structure was decreased.However,the protein secondary structure was stable under the 600 mg/L lead stress for the whole plant.The result indicated that sunflower HA300 seedlings could maintain the stability of their protein secondary structure under a certain concentration of lead stress.Our study provided a scientific basis for the study of physiological mechanisms of sunflower seedlings under stress.
