基于土壤宏基因组的新颖木聚糖酶基因克隆及生物信息学分析

摘　　要：土壤微生物总DNA经试剂盒提取,使用CODEHOP引物进行木聚糖酶基因编码区核心序列进行扩增,根据所扩增序列设计hiTAIL-PCR特异性引物,最终克隆了1个编码区长度为1284 bp的新颖木聚糖酶基因。生物信息学分析表明,该基因编码蛋白质氨基酸序列长度为427 aa,分子量为47398.91 Da,理论等电点(pI)为8.99,是一个不含跨膜区域的亲水性蛋白。The total DNA of soil microorganism was extracted by kit,and the core sequence of xylanase gene coding region was amplified by CODEHOP primer.According to the amplified sequence,the specific primers of hiTAIL-PCR was designed.Finally,a novel xylanase gene with 1284 bp coding region was cloned.Bioinformatics analysis showed that the amino acid sequence of the gene was 427 aa,the molecular weight was 47398.91 Da,the theoretical isoelectric point(pI)was 8.99,and it was a hydrophilic protein without transmembrane region.
