摘 要:目的探讨天然药物姜黄素及其衍生物FM0807对鼠源性肝癌H22细胞增殖抑制作用以及对负性免疫调节因子PD-L1表达影响,分析二者之间的相关性。方法以体外培养的H22细胞为研究对象,考察姜黄素及衍生物对H22细胞的增殖抑制作用;姜黄素及FM0807单独给药或者联合γ-干扰素处理后,流式细胞术检测H22细胞PD-L1表达的变化;结果姜黄素及其衍生物对H22增殖抑制作用呈浓度依赖性;姜黄素对H22细胞PD-L1的诱导表达作用呈时间和浓度依赖性;而衍生物FM0807对PD-L1的诱导表达作用则仅表现为浓度依赖性而不具有时间依赖性;两药均可协同IFN-γ诱导PD-L1表达。结论姜黄素及其衍生物对H22增殖抑制作用与PD-L1表达呈正相关;衍生物FM0807较母药姜黄素不易诱导免疫耐受,具有一定的开发价值。OBJECTIVE To investigate the effects of Curcumin and its derivative FM0807 on proliferation and PD-L1 expression in Hepatic Cancer Cell Line H22,and analyze the correlation between the two.METHODS Mice hepatocyte cell line H22 cultured in vitro was treated with Curcumin or FM0807,cell proliferation activity was determined by MTT assay.Curcumin and FM0807 were administered alone or in combination with gamma-interferon,and the effect of different treatment on the expression of PD-L1 was investigated by flow cytometry in vitro.RESULTS Curcumin and FM0807 significantly inhibited the proliferation of H22 cells with a dose-dependent manner at low concentrations.Curcumin could up-regulate the expression of PD-L1 in H22 cells with a dose-and time-dependent manner,while FM0807 just with a dose-dependent manner during 24 h.In addition,Curcumin and its derivative potentiated IFN-γ-induced PD-L1 expression in H22 cells.CONCLUSION The inhibitory effect of Curcumin and its derivative FM0807 on H22 proliferation was positively correlated with PD-L1 expression.The derivative FM0807 is not easy to induce immune tolerance and has certain development value relative to the parent drug Curcumin.
