摘  要:目的建立小分子量透明质酸片段的分子量及其分布的测定方法。方法采用高效凝胶渗透色谱法(HPGPC)、琼脂糖凝胶电泳法、凝胶渗透色谱与多角度激光散射联用(GPC-MALLS)以及特性粘度法对透明质酸注射液的分子量进行测定。结果GPC-MALLS法得到的分子量变异系数小于5%,测定结果精确度较高,分布宽度Mw/Mn较小;HPGPC法得到的分子量偏低且分布宽度Mw/Mn较大,但结果的变化趋势与GPC-MALLS法及电泳的直观结果相一致;特性粘度法对于介于10~100kDa分子量范围内的样品,测得的数值差异较大。结论GPC-MALLS结合凝胶电泳法是最好的测定小分子透明质酸片段的分子量和分布范围的方法。OBJECTIVE Measurement of molecular weight and molecular distribution of low molecular hyaluronic acid fragments.METHODS HPGPC,agarose gel electrophoresis,GPC-MALLS and intrinsic viscosity method were employed for comparative study.RESULTS CV of measurement by using GPC-MALLS is less than 5%.The distribution width of Mw/Mn measured by GPC-MALLS was relatively narrow,indicating that GPC-MALLS was a more precise method for the measurement.The molecular weight measured by HPGPC was relatively lower than those measured by GPC-MALLS and agarose gel electrophoresis.The distribution width of Mw/Mn measured by HPGPC was wider than that measured by GPC-MALLS.The results obtained by HPGPC more or less were close to those obtained by GPC-MALLS and agarose gel electrophoresis.The molecular weight results measured by the intrinsic viscosity were very different from those obtained by GPC-MALLS and agarose gel lectrophoresis,indicating that the intrinsic viscosity method was not a proper method for measuring molecular weight of hyaluronic acid fragments at range of 10-100 kDa molecular weight.CONCLUSION Combined use of GPC-MALLS and agarose gel electrophoresis is the best method for measurement of molecular weight and molecular distribution of low molecular hyaluronic acid fragments at range of 10-100 kDa.
