摘 要:目的观察转移相关蛋白2(MTA2)、低氧诱导因子1α(HIF-1α)和上皮钙黏蛋白(E-cadherin)在喉鳞状细胞癌(简称喉癌)组织及转染MTA2siRNA喉癌细胞中的表达变化,并探讨其临床意义。方法分别采用免疫组织化学S-P法及Western blotting法检测喉癌组织及癌旁正常喉组织中MTA2、HIF-1α、E-cadherin,分析MTA2、HIF-1α、E-cadherin与患者临床病理特征的关系及三者之间的相关性;使用脂质体Lipofectamine 2000将MTA2siRNA导入喉癌细胞系Hep-2中,观察HIF-1α、E-cadherin的表达变化。结果喉癌及癌旁正常喉组织中MTA2蛋白阳性细胞率分别为69.1%、14.3%,HIF-1α蛋白阳性细胞率分别为74.5%、11.4%,E-cadherin蛋白阳性细胞率分别为38.2%、91.4%,两种组织中MTA2、HIF-1α、E-cadherin蛋白阳性细胞率比较,P均<0.05。喉癌及癌旁正常喉组织中MTA2蛋白相对表达量分别为0.95±0.10、0.73±0.07,HIF-1α分别为1.11±0.11、0.56±0.08,E-cadherin分别为0.98±0.12、1.31±0.09,两种组织中MTA2、HIF-1α、E-cadherin蛋白相对表达量比较,P均<0.05。喉癌组织中MTA2和HIF-1α表达均与喉癌组织学分级、临床分期和淋巴结转移相关(P均<0.05);E-cadherin表达与喉癌临床分期和淋巴结转移相关(P均<0.05)。喉癌组织中,MTA2与HIF-1α蛋白表达呈正相关(r=0.512,P=0.000),与Ecadherin蛋白表达呈负相关(r=-0.608,P=0.000)。喉癌细胞系Hep-2中干扰MTA2蛋白的表达后,HIF-1α蛋白表达下调,E-cadherin蛋白表达上调。结论喉癌组织中MTA2和HIF-1α高表达,E-cadherin低表达;转染MTA2siRNA喉癌细胞中HIF-1α蛋白表达下调,E-cadherin蛋白表达上调;MTA2、HIF-1α、E-cadherin蛋白可能与喉癌的发生及转移有关,且MTA2和E-cadherin蛋白之间具有协同作用,MTA2与HIF-1α蛋白之间具有拮抗作用。Objective To investigate the expression changes of metastasis-associated protein-2(MTA2),hypoxia inducible factor-1α(HIF-1α)and Epithelial-cadherin(E-Cadherin)in the laryngeal squamous cell carcinoma(LSCC)tissues and laryngeal carcinoma cells transfected with MTA2 siRNA,and to discuss their clinical significance.Methods The expression levels of MTA2,HIF-1α,and E-Cadherin in the laryngeal cancer tissues and adjacent normal laryngeal tissues were determined by immunohistochemical S-P method,and the relationship between their expression and the clinical pathological features and the correlations among the three proteins were analyzed.Lipofectamine 2000 was used to introduce MTA2 siRNA into Hep-2 cell to observe the expression changes of HIF-1 and E-cadherin.Results In the laryngeal cancer and adjacent normal laryngeal tissues,the positive rates of MTA2,HIF-1 and E-Cadherin were 69.1%and 14.3%,74.5%and 11.4%,38.2%and 91.4%,respectively;the relative expression levels of MTA2,HIF-1 and E-Cadherin were 0.95±0.10 and 0.73±0.07,1.11±0.11 and 0.56±0.08,and 0.98±0.12 and 1.31±0.09,respectively;significant differences were found in the positive rates and relative expression of MTA2,HIF-1 and E-Cadherin proteins between these two tissues(all P<0.05).The expression of MTA2 and HIF-1αwas related to histological grade,clinical stage,and lymph node metastasis(all P<0.05),the expression of E-Cadherin was related to the clinical stage and lymph node metastasis(both P<0.05).The MTA2 expression was positively correlated with HIF-1αexpression(r=0.512,P=0.000)and was negatively correlated with E-Cadherin(r=-0.608,P=0.000)in the LSCC tissues.After MTA2 protein was restrained,the expression of HIF-1αprotein was down-regulated and E-Cadherin protein was up-regulated in Hep-2 cells.Conclusions Both MTA2 and HIF-1αare highly expressed and HIF-1αis low expressed in the LSCC tissues,and the expression of HIF-1αprotein is down-regulated and the expression of E-Cadherin protein is up-regulated in laryngeal carcinoma cells transfe
