摘  要:目的:探讨血栓形成性疾病患者F2R样凝血酶/胰蛋白酶受体3(F2RL3)基因甲基化水平,以评价F2RL3基因甲基化鉴别血栓性疾病的效能。方法:收集246例血栓性患者的EDTA-K2抗凝全血标本,包括颈内静脉血栓(IJVT)37例、下腔静脉栓塞(IVCT)49例、下肢静脉栓塞(LEVT)32例、心肌梗死(MI)52例、脑梗死(CI)76例和健康对照组(Ctrl)76例,采用实时荧光定量PCR技术检测全血F2RL3基因启动子甲基化水平,并分析其与危险因素的相关性。结果:①246例血栓性疾病患者全血F2RL3基因甲基化水平为(0.49±0.11),低于Ctrl组(0.60±0.15),差异有统计学意义(P<0.05)。②静脉血栓患者(VT组)全血F2RL3基因甲基化水平为(0.50±0.08),动脉血栓者(AT组)F2RL3基因甲基化水平为(0.49±0.12),均低于Ctrl组(P<0.05)。③IVCT组患者(0.48±0.08)、LEVT组患者(0.49±0.11)和CI患者(0.48±0.09)F2RL3基因甲基化水平低于Ctrl组,差异有统计学意义(P<0.05)。结论:全血F2RL3基因甲基化水平降低可辅助诊断血栓形成性疾病。Objective:To analyze the methylation level of F2 RL3(coagulation factor II thrombin receptor-like 3)gene in patients with various thrombotic diseases.Method:EDTA-2 K anticoagulated whole blood were collected of total of 246 patients with thrombotic disease,including 37 cases of internal jugular vein thrombosis(IJVT),49 cases of inferior vena cava thrombosis(IVCT),32 cases of lower extremity venous thrombosis(LEVT),52 cases of myocardial infarction(MI),and 76 cases of cerebral infarction(CI)and 76 healthy controls(Ctrl).Real-time fluorescent quantitative PCR(RT-PCR)was used to detect the whole blood F2 RL3 gene promoter methylation level.Result:In 246 patients with thrombotic disease,the F2 RL3gene methylation level was decreased in whole blood samples,and the methylation level of F2 RL3gene in venous thrombosis and arterial thrombosis was decreased.Conclusion:The reduction of F2 RL3methylation level in whole blood can assist in the diagnosis of thrombotic diseases.
