摘 要:目的探讨长链非编码RNA(LncRNA)在喉癌患者血清、癌组织中的表达及其与预后的关系。方法选取本院2013年1月~2018年1月确诊的喉癌患者100例为观察组,选取年龄、性别与观察组匹配的健康体检者60例为对照组,通过实时荧光定量PCR(qRT-PCR)测定两组血清LncRNA H19表达水平;癌组织、配对癌旁组织经手术切除获取,检测患者血清及癌组织、配对癌旁组织LncRNA H19表达水平,根据癌组织、癌旁组织及血清LncRNAH19表达水平将观察组分为高表达组(55例)与低表达组(45例),并分析癌组织LncRNA H19表达水平与患者病理特征及预后的关系。结果观察组血清LncRNA H19相对表达水平显著高于对照组(P<0.05);癌组织LncRNA H19相对表达水平显著高于配对癌旁组织(P<0.05);观察组血清与癌组织LncRNA H19相对表达水平显著正相关(P<0.05);LncRNA H19高表达组TNM分期III期、低分化、淋巴结转移比例均显著高于低表达组(P<0.05);LncRNA H19高表达组患者总生存时间、无进展生存时间均显著短于低表达组(P<0.05)。结论喉癌患者血清、癌组织LncRNA H19表达均上调,LncRNA可能参与喉癌发生、进展过程,可作为喉癌患者预后评估的有效指标。Objective To investigate the expression of long-chain non coding RNA(LncRNA) in serum and cancer tissues of patients with laryngeal cancer and its relationship with prognosis. Methods 100 patients with laryngeal cancer diagnosed in our hospital from January 2013 to January 2018 were selected as the observation group. 60 healthy persons matched with age and sex in the observation group were selected as the control group. The expression level of serum LncRNA H19 in the two groups was determined by real-time fluorescent quantitative PCR(QRT PCR). The expression of lncrna H19 in serum, cancer tissue and matched cancer tissue were detected. The observation group was divided into high expression group(55 cases) and low expression group(45 cases) according to the expression level of lncrnah19 in cancer tissue, paracancerous tissue and serum. The relationship between the expression level of lncrnah19 in cancer tissue and the pathological characteristics and prognosis of patients was analyzed. Results The relative expression level of serum LncRNA 19 in the observation group was significantly higher than that in the control group(P<0.05). The relative expression level of LncRNA H19 in cancer tissues was significantly higher than that in matched adjacent tissues(P<0.05). There was a significant positive correlation between the relative expression level of LncRNA H19 in serum and cancer tissues in the observation group(P<0.05). The proportion of TNM stage III, low differentiation and lymph node metastasis in the high expression group was significantly higher than that in the low expression group(P<0.05). The total survival time and progression free survival time of patients in the high expression group of LncRNA H19 were significantly shorter than those in the low expression group(P<0.05). Conclusion The expression of LncRNA H19 in the serum and tissues of patients with laryngeal cancer is up-regulated. LncRNA may be involved in the process of carcinogenesis and progression of laryngeal cancer, which can be used as an effect
