摘 要:目的探索碘过量对小鼠胰岛β细胞形态的影响。方法将60只体重为(18~22)g/只的雌性(BALB/c)小鼠随机分为6组,每组10只,在饮水中加入不同浓度的碘,对照组给予正常浓度碘(5μg/L)的自来水。碘过量的暴露组采用在饮用水中加入碘酸钾的方法,给予分别含有300μg/L、600μg/L、1200μg/L、2400μg/L和4800μg/L碘的饮用水。饲养1个月,取小鼠胰腺组织,光镜和电镜下观察胰岛β细胞形态。结果对照组小鼠胰岛β细胞形态结构正常,呈椭圆形,与周围细胞界限清楚。高浓度碘水饲养小鼠1个月后,光镜下可见,从B组(300μg/L)开始胰岛β细胞形态发生改变,细胞体积增大,胰岛增生、肥大,胰岛β细胞肿胀、形态不规则。在电镜下可见,从D组(1200μg/L)开始细胞超微结构发生明显变化,线粒体数量减少,出现肿胀、变性,胰岛β细胞肿胀、纤维化和变性,甚至出现缺如,内质网扩张、增粗,线粒体嵴向基底部聚集甚至断裂或呈空泡状等。结论碘过量对小鼠胰岛β细胞形态产生影响,可以损伤小鼠胰岛β细胞。Objective Exploring the effect of iodine excess on the morphology of mouse isletβcells.Methods Sixty females(BALB/c)weighing 18-22 g/only were randomly divided into 6 groups,10 in each group,and different concentrations of iodine were added to the drinking water.The control group was given normal concentration of iodine(5μg/L)tap water.The iodine excess exposure group was treated with potassium iodate in drinking water,and drinking water containing 300μg/L,600μg/L,1200μg/L,2400μg/L,and 4800μg/L iodine,respectively,was administered.After 1 month of feeding,the pancreatic tissue of the mice was taken,and the morphology ofβislet cells was observed by light microscopy and electron microscope.Results In the control group,the islet β cells had normal morphological structure and were elliptical.There had a clear boundaries with surrounding cells.After 1 month of high concentration iodine water feeding,from group B(300μg/L),light microscopy showed that the isletβcells morphology changed,the cell volume increased,islet hyperplasia,hypertrophy,isletβcell swelling,and morphology rule.Under electron microscope,the ultrastructure of cells began to change significantly from group D(1200μg/L).The number of mitochondria is reduced,swelling,degeneration,swelling of isletβcells,fibrosis and degeneration,and even absent,endoplasmic reticulum expansion,thickening,mitochondria sputum aggregation or even rupture or vacuolization.Conclusion Excessive iodine affects the morphology of mouse isletβcells and can damage mouse isletβcells.
