沉默Pax3表达对神经胶质瘤细胞增殖、侵袭和迁移能力的影响

摘　　要：目的探讨沉默配对盒基因3(Pax3)表达对神经胶质瘤细胞增殖、侵袭和迁移能力的影响。方法培养神经胶质瘤细胞系U87,分为三组:Pax3沉默组,将Pax3小干扰RNA转染到U87细胞中;阴性对照组,将无义寡聚核苷酸转染到U87细胞中;空白对照组,加入PBS 20μL。检测三组Pax3 mRNA的表达水平,观察细胞增殖、侵袭及迁移能力的变化。结果空白对照组、阴性对照组和Pax3沉默组Pax3 mRNA表达量分别为0.81±0.12、0.79±0.03和0.15±0.07;其中,Pax3沉默组Pax3 mRNA表达水平低于空白对照组和阴性对照组(P<0.05)。Pax3沉默组细胞增殖能力较空白对照组和阴性对照组降低(P<0.05)。Pax3沉默组细胞侵袭和迁移能力低于空白对照组及阴性对照组(P<0.05)。结论沉默Pax3表达能够抑制神经胶质瘤细胞增殖、侵袭和迁移能力,Pax3可能参与了神经胶质瘤的发生、发展。Objective To investigate the effects of silencing paired box gene 3(Pax3) expression on the proliferation,invasion and migration of the glioma cells.Methods The glioma cell line U87 was cultured and divided into three groups.Group A was transferred with Pax3 siRNA,group B was transferred with senseless oligodeoxynucleotides and group C was treated with PBS 20 μL.The mRNA expression of Pax3 was detected and the cell proliferation,invasion and migration were observed.Results The mRNA expressions of groups of C,B and A were 0.81±0.12,0.79±0.03 and 0.15±0.07,respectively,which were lower in group A than those in groups of B and C(P<0.05).The ability of cell proliferation,invasion and migration was lower in group A than that in groups of B and C(P<0.05).Conclusion Silencing Pax3 expression can inhibit the ability of cell proliferation,invasion and migration of glioma cells.Pax3 may be involved in the occurrence and development of glioma cells.
