摘 要:目的探讨新生淋巴管及肿瘤内趋化因子受体4(CXCR4)和趋化因子受体7(CCR7)在卵巢癌淋巴转移中的作用机理。方法通过腹腔注射人卵巢腺癌细胞SKOV3建立荷人卵巢癌细胞裸鼠腹腔移植瘤模型后随机分为两组,处理组(10只)腹腔注射趋化因子抑制剂AMD3100 0.01 mg/g,对照组(10只)腹腔注射PBS 0.2 mL。采用免疫组化染色方法检测两组卵巢癌移植瘤、腹腔淋巴结和皮下淋巴结中基质细胞衍生因子(SDF-1)、次级淋巴组织细胞趋化因子(CCL21)、CXCR4和CCR7表达量。采用PCR检测三种不同组织中VEGF-C和VEGF-D mRNA的表达。结果VEGF-C和VEGF-D mRNA在两组卵巢癌移植瘤中均高表达,且其在处理组中的表达低于对照组(P<0.05)。CXCR4和CCR7在两组卵巢癌移植瘤中的表达高于皮下淋巴结和腹腔淋巴结(P<0.05),且其在处理组中的表达低于对照组(P<0.05)。SDF-1和CCL21在两组卵巢癌移植瘤中的表达低于皮下淋巴结和腹腔淋巴结(P<0.05),且其在处理组中的表达低于对照组(P<0.05)。结论肿瘤细胞在CXCR4和CCR7的作用下向淋巴管趋动,新生淋巴管与组织淋巴管交互,从而使肿瘤细胞沿淋巴管道向淋巴结定向归巢。Objective To explore the mechanism of CXCR4 and CCR7 in neonatal lymphoid vessels and tumors in lymphatic metastasis of oval cancer.Methods Abdominal transplantation model of human ovarian cancer cells in nude mice was established in 20 nude mice by intraabdominal injection of SKOV3 cells,which were randomly divided into two groups with 10 mice each.The mice in group A were treated with intraabdominal injection of chemotactic factor inhibitor AMD3100 0.01 mg/g and those in group B were treated with intraabdominal injection of PBS 0.2 ml as the controls.The expressions of SDF-1,CCL21,CXCR4 and CCR7 in the xenografts,abdominal lymph node and subcutaneous lymph node were detected using immunohistochemical method.The expressions of VEGF-C and VEGF-D mRNA in the xenografts,abdominal lymph node and subcutaneous lymph node were detected using PCR technique.Results VEGF-C and VEGF-D mRNA were highly expressed in the xenografts of two groups,which were lower in group A than those in group B(P<0.05).The expressions of CXCR4 and CCR7 in the xenografts of two groups were higher than those in abdominal lymph node and subcutaneous lymph node(P<0.05),which were higher in group A than those in group B(P<0.05).The expressions of SDF-1 and CCL21 in the xenografts of two groups were lower than those in abdominal lymph node and subcutaneous lymph node(P<0.05),which were lower in group A than those in group B(P<0.05).Conclusion Under the action of CXCR4 and CCR7,the tumor cells tend to move to the lymphatic vessels,and the new lymphatic vessels interact with the lymphoid vessels of the tissues,so that the tumor cells homing to the lymph nodes along the lymphoid duct.
