摘 要:目的分别探讨Fra-1(fos-related antigen-1)、Fra-2(fos-related antigen-2)基因在喉鳞状细胞癌(laryngeal squamous carcinoma cell, LSCC)中表达情况及两者的相关性。方法采用免疫组化SP法及逆转录-多聚酶链反应(reverse transcription-polymerase chain reaction, RT-PCR)检测手术切除标本47例LSCC组织和21例癌旁正常组织中的Fra-1和Fra-2基因在蛋白及mRNA水平的表达情况,在癌组织中分析两者的表达与各临床病理参数的关系,进一步探讨两基因表达的相关性。结果 Fra-1和Fra-2基因在LSCC中蛋白水平阳性表达率均高于癌旁正常组织(P均<0.05),LSCC中两基因蛋白水平表达与临床分期、淋巴结转移、吸烟相关,与病理分级、年龄、解剖分区无关;两者mRNA水平表达量均高于癌旁正常组织(P均<0.01),Fra-1基因表达情况受到临床分期、病理分级、淋巴结转移情况以及吸烟参数影响,与年龄、解剖分区参数不相关。而Fra-2的表达仅与病理分级相关,与临床分期、淋巴结转移以及吸烟、年龄、解剖分区等参数不相关。此外Fra-1、Fra-2基因在mRNA水平表达量数值行直线相关分析呈正相关(r=0.570,P<0.01)。结论 LSCC组织中Fra-1、Fra-2基因在蛋白及mRNA水平表达均增高,两者的表达产物呈正相关。两基因之间可能相互活化表达促进喉癌的发生与发展。Objective To investigate the expression of Fra-1(Fos-related antigen-1) and Fra-2(Fos-related antigen-2) in laryngeal squamous carcinoma cell( LSCC), as well as their correlation.Methods Protein expression and mRNA expression of Fra-1 and Fra-2 were detected by SP immunohistochemistry and reverse transcription-polymerase chain reaction(RT-PCR) in 47 LSCC tissues and 21 normal adjacent tissues. The relations between the expression of the two genes as well as those between them and clinicopathological parameters in tumor tissue were analyzed.Results The positive expression rates of Fra-1 and Fra-2 protein in LSCC were both higher than those in the normal adjacent tissues(both P<0.05). Their protein expression levels in LSCC were associated with clinical stage, lymph node metastasis and smoking, but not associated with pathological grade and anatomic site of tumor, or patients’ age. The expression levels of mRNA of Fra-1 and Fra-2 in LSCC were higher than those in the normal adjacent tissues(both P<0.01). The mRNA expression of Fra-1 was related to clinical stage, pathological grade, lymph node metastasis and smoking, but not related to patients’ age and anatomic site of tumor. The mRNA expression of Fra-2 was not associated with all the parameters except for pathological grade. In addition, linear correlation analysis revealed positive correlation between the mRNA expression of Fra-1 and Fra-2(r=0.570, P<0.01).Conclusions The expression of Fra-1 and Fra-2 in LSCC is increased at both mRNA and protein levels, and the expression products of Fra-1 and Fra-2 are positively correlated. The two genes may activate with each other to promote the development and progression of LSCC.
