摘  要:超高压液相色谱法(UPLC)是一种新型液相色谱技术,采用小粒径色谱填料和超高压系统,与高效液相色谱法(HPLC)相比,UPLC具有柱效更高、分析速度更快,以及灵敏度更高的优点,已在许多领域广泛应用。该文设计了超高压液相色谱法快速分离咖啡因、阿司匹林和非那西丁,以及外标法定量测定复方阿司匹林片中咖啡因含量的实验,介绍了超高压液相色谱法的基本原理、仪器组成和操作、优化色谱条件以及外标定量的方法,可进一步加深学生的色谱理论知识,有效提高学生对大型仪器UPLC的基本操作能力,并锻炼学生的科研思维能力。Ultra high performance liquid chromatography(UPLC)is a novel technique in liquid chromatography.It is more efficient,rapid and sensitive than high performance liquid chromatography(HPLC),owing to using smaller particle size in analytical column.Because of these advantages,UPLC has been widely used in many fields.Experiments on rapid separation of caffeine,aspirin and phenacetin by UPLC and quantitative analysis of caffeine in compound aspirin tablets by external standard method were designed.In the experimental teaching,the basic principle,instrument composition and operation,the general methods of optimizing chromatographic conditions and the external standard method were introduced.Through this experiment,students not only better understood the theoretical knowledge,but also learned the operation of UPLC.More importantly,students5 experimental skills have been improved,their scientific thinking ability has been exercised.
