摘  要:目的:研究cAMP-PKA信号通路参与慢性心理应激大鼠心律失常的机制。方法:将60只SD大鼠随机分为空白组、模型对照组和干预组,每组各有20只大鼠。采用Cronli法对模型对照组大鼠和干预组大鼠进行干预,建立慢性抑郁大鼠模型,并用氟西汀为干预组大鼠灌胃。建模及干预后,观察并比较三组大鼠心律失常的发生率及其心肌中环磷酸腺苷(cAMP)的水平及蛋白激酶A(PKA)的密度值。结果:建模及干预后,模型对照组大鼠与干预组大鼠室性早搏的发生率、房性早搏的发生率和房室交接性早搏的发生率均高于空白组大鼠,P<0.05;干预组大鼠室性早搏的发生率低于模型对照组大鼠,P<0.05;模型对照组大鼠与干预组大鼠房性早搏的发生率及房室交接性早搏的发生率相比,P>0.05。建模及干预后,模型对照组大鼠心肌中cAMP的水平及PKA的密度值均低于空白组大鼠与干预组大鼠,P<0.05;空白组大鼠与干预组大鼠心肌中cAMP的水平及PKA的密度值相比,P>0.05。结论:心理应激大鼠发生心律失常与其cAMP-PKA信号通路下调有关,氟西汀可通过上调心理应激大鼠心肌中cAMP和PKA的表达而起到缓解其心律失常的作用。objective:to study the mechanism of camp-pka signaling pathway involved in arrhythmia in rats with chronic psychological stress.Methods:60 SD rats were randomly divided into blank group,model control group and intervention group,with 30 rats in each group.Cronli method was used to intervene the rats in the model control group and the intervention group,to establish the rat model of chronic depression,and the rats in the intervention group were gavaged with fluoxetine.After modeling and intervention,the incidence of arrhythmia,the level of cAMP and the density of protein kinase A(PKA)were observed and compared.Results:after modeling and intervention,the incidence of ventricular premature beats,atrial premature beats and atrioventricular handover premature beats in the model control group and the intervention group were higher than those in the blank group(P<0.05).The incidence of ventricular premature beat in the intervention group was lower than that in the model control group(P<0.05).Compared with the intervention group,the incidence of atrial premature beats and the incidence of atrioventricular handover premature beats in the model control group was P>.05.After modeling and intervention,the level of cAMP and the density of PKA in the model control group were lower than those in the blank group and the intervention group(P<0.05).The level of cAMP and the density of PKA in the myocardium of rats in the blank group were compared with that in the intervention group(P>,0.05).Conclusion:arrhythmia in psychological stress rats is related to the down-regulation of camp-pka signaling pathway.Fluoxetine can alleviate arrhythmia by up-regulating the expression of cAMP and PKA in the myocardium of psychological stress rats.
