摘 要:目的在细胞水平探讨Beclin1在CO2气腹促进人卵巢癌细胞SKOV3增殖转移过程中的作用。方法采用脂质体介导的瞬时转染法转染,分成Beclin1过表达组、对应的GV362空质粒组、Beclin1干扰表达组、对应的pGPH1空质粒组、单纯脂质体组(空白对照组),CO2压力相同,作用相同时间。采用qRT-PCR(定量反转录聚合酶连锁反应)和Western blot检测各组细胞中Beclin1的mRNA表达情况,采用超氧化物歧化酶法(WST-1)检测各组细胞的增殖情况,采用Transwell实验检测各组细胞的迁移能力。结果Beclin1过表达组的卵巢癌SKOV3细胞增殖率明显升高,干扰质粒组细胞增值率明显降低(P<0.05)。Beclin1过表达组穿透进入下室的SKOV3细胞明显增多,而Beclin1干扰表达质粒组则明显减少(P<0.05)。结论Beclin1在CO2气腹中能促进人卵巢癌细胞株SKOV3的增殖与迁移。Objective To investigate the role of Beclin1 in enhancing the proliferation and metastasis of human ovarian cancer cell line SKOV3 in the course of carbon dioxide pneumoperitoneum.Methods Liposome-mediated transient transfection was used,and there were Beclin1 Overexpression Group,corresponding GV362 Empty Plasmid Group,Beclin1 Interference Expression Group,corresponding pGPH1 Empty Plasmid Group and Simple Liposome Group(control group)divided with the same CO2 pressure and action time.qRT-PCR and Western blot were used to detect the mRNA expression of Beclin1 in the cells in each group.WST-1 was used to detect the proliferation of the cells in each group.Transwell assay was used to detect the cell migration ability in each group.Results The Beclin1 Overexpression Group had the proliferation rate of SKOV3 significantly increased and had the Beclin1 Interference Expression Group had the proliferation rate significantly decreased(P<0.05).The Beclin1 Overexpression Group had the number of SKOV3 cells penetrating into the lower chamber significantly increased while the Beclin1 Interference Expression Group had such number significantly decreased(P<0.05).Conclusion Beclin1 can enhance the proliferation and migration ability of ovarian cancer SKOV3 cells in CO2 pneumoperitoneum.
