摘 要:目的探讨中药肠复方及拆方的抑瘤作用及其对结肠癌荷瘤小鼠不同组织中髓源抑制性细胞(MDSCs)、CD4^+、CD8^+T细胞的影响。方法建立结肠癌细胞株CT26荷瘤小鼠模型,随机分为5组,各8只。分别给予肠复方全方、拆方Ⅰ(健脾益气扶正)、拆方Ⅱ(祛湿化瘀解毒)、香菇多糖、蒸馏水(对照组)进行干预,比较各组荷瘤小鼠的抑瘤率,采用流式细胞仪检测小鼠脾脏细胞悬液、外周血、肿瘤细胞悬液中MDSCs水平及外周血、肿瘤细胞悬液中CD4^+、CD8^+T细胞水平。结果肠复方全方组、拆方Ⅰ组、拆方Ⅱ组、香菇多糖组荷瘤小鼠的肿瘤生长均受到抑制,抑瘤率分别为30.1%、16.1%、26.7%、13.8%;脾重抑制率分别为19.5%、9.6%、15.5%、5.6%。肠复方全方组、拆方Ⅰ组、拆方Ⅱ组、香菇多糖组外周血MDSCs水平均显著低于对照组(P<0.05),且全方组<拆方Ⅱ组<拆方Ⅰ组<香菇多糖组(P<0.05);肠复方全方组、拆方Ⅰ组、拆方Ⅱ组脾脏中MDSCs水平均显著低于对照组(P<0.01),且全方组<拆方Ⅱ组<拆方Ⅰ组(P<0.05),而香菇多糖组脾脏中MDSCs水平与对照组无明显差异(P>0.05);全方组、拆方Ⅱ组肿瘤组织中MDSCs水平均显著低于对照组(P<0.05),而拆方Ⅰ组、香菇多糖组肿瘤组织中MDSCs水平低于对照组,但差异无统计学意义(P>0.05)。与对照组相比,肠复方全方组、拆方Ⅰ组、拆方Ⅱ组、香菇多糖组外周血和肿瘤组织中CD4^+、CD8^+T细胞水平均显著升高(P<0.05),且全方组>拆方Ⅰ组>拆方Ⅱ组>香菇多糖组(P<0.01)。结论肠复方及其拆方均可抑制结肠癌荷瘤小鼠的肿瘤生长,健脾益气扶正组分(拆方Ⅰ)和祛湿化瘵解毒组分(拆方Ⅱ)在提高机体免疫状态、抑制肿瘤发生发展方面各有侧重,二者协同可使全方发挥最佳疗效,其机制可能与抑制MDSCs的增殖及提高T细胞水平有关。Objective To investigate the anti-tumor effect of Changfufang and its decomposed recipes and its effects on myeloidderived suppressor cells(MDSCs),CD4^+and CD8^+T cells in different tissues of colorectal cancer-bearing mice.Methods The model of CT26 tumor-bearing mice was established and randomly divided into five groups,with eight mice in each group.They were respectively intervened with Changfufang,decomposed recipeⅠ(Jianpi Yiqi Fuzheng),decomposed recipeⅡ(Qushi Huayu Jiedu),lentinan and distilled water(control group).After the intervention,the tumor inhibition rate of each group was calculated.And the levels of MDSCs in splenic cell suspension,peripheral blood and tumor cell suspension,and the levels of CD4^+and CD8^+T cells in peripheral blood and tumor cell suspension were detected by flow cytometry.Results The tumor growth was inhibited in Changfufang group,decomposed recipeⅠgroup,decomposed recipeⅡgroup,and lentinan group,and the inhibition rate was 30.1%,16.1%,26.7%and 13.8%,respectively.The inhibition rate of the spleen was 19.5%,9.6%,15.5%and 5.6%respectively.The level of MDSCs in peripheral blood was in a order from low to high in the Changfufang group,the decomposed recipeⅠgroup,the decomposed recipeⅡgroup and the lentinan group(P<0.05).Moreover,the levels of MDSCs in peripheral blood in the above groups were all lower than that in control group(P<0.01).In the spleen of the tumorbearing mice,the level of MDSCs was also in a order from low to high in the Changfufang group,the decomposed recipeⅠgroup,and the decomposed recipeⅡgroup(P<0.05),and they were all lower than that in control group(P<0.01).But no obvious difference was found in MDSCs levels in the spleen between the lentinan group and the control group(P>0.05).In tumor tissues,only the Changfufang group and the decomposed recipeⅡgroup showed an obvious inhibition effect on the MDSCs(P<0.05).The MDSCs levels in the decomposed recipeⅠand the lentinan was also decreased,but the difference had no statistical significance(P>0.05).
