摘 要:目的探讨丹参川芎嗪对链脲佐菌素(STZ)诱发的糖尿病大鼠神经病理性疼痛的改善效果。方法选取SPF级Sprague Dawley大鼠,腹腔注射STZ,72 h后进行血糖测定,筛选符合标准的糖尿病大鼠。将30只大鼠等分为3组,每组10只,分别为空白对照组(Control组)、糖尿病组(DM组)、糖尿病协同丹参川芎嗪治疗组(DM+SML组)。对DM+SML组大鼠每天腹腔注射丹参川芎嗪干预(浓度为2 mL/kg),连续治疗8周,分别于丹参川芎嗪注射的第0、2、4、6和8周对全部大鼠进行机械痛阈值、热痛阈值和神经传导速率检测,并对大鼠脊髓p38MAPK和IL-1β的基因表达进行定量。结果丹参川芎嗪治疗的2、4、6、8周,DM+SML组大鼠的足底机械痛阈值和热痛阈值高于DM组(P均<0.05),且DM+SML组大鼠的运动神经传导速率高于DM组(P均<0.05)。同时,DM+SML组大鼠的脊髓p38MAPK和IL-1β基因表达低于DM组(P均<0.05)。结论丹参川芎嗪可有效改善糖尿病大鼠的神经病理性疼痛。Objective To investigate the effect of salviae miltiorrhizae and ligustrazine(SML)on neuropathic pain induced by streptozotocin(STZ)in diabetic rats.Methods The SPF Sprague-Dawley rats were intraperitoneally injected with STZ for 72 h,and the glucose levels were determined to obtain the qualified diabetic rats.The rats were then divided into three groups,including the blank control group(Control group),the diabetes group(DM group),and the diabetes+SML group(DM+SML group),with 10 rats in each group.The rats in the DM+SML group were injected with SML per day(2 mL/kg)for 8 weeks.The mechanical pain threshold,thermal pain threshold,and nerve conduction rate of all rats were measured at week 0,2,4,6 and 8 after injection of SML,and the gene expression levels of p38MAPK and IL-1βin the spinal cord of rats were quantified.Results At the 2nd,4th,6th and 8th weeks after treatment,the plantar mechanical pain threshold and thermal pain threshold in the DM+SML group were significantly higher than those in the DM group(both P<0.05),and the motor nerve conduction velocity in the DM+SML group was higher than that in the DM group(P<0.05).Moreover,the expression levels of p38MAPK and IL-1βin the spinal cord of DM+SML group were lower than those of the DM group(both P<0.05).Conclusion SML can effectively improve neuropathic pain in diabetic rats.
