摘 要:目的探讨胶质瘤组织和胶质瘤干细胞(CSCs)miR-520f-3p表达及临床意义。方法采用qRT-PCR法检测miR-520f-3p在40例份胶质瘤组织和20例份脑组织中的表达,并分析胶质瘤组织中miR-520f-3p表达与患者临床病理特征的关系。磁珠分选胶质瘤细胞系U87中的CSCs。Western blotting法检测OCT4、Nanog、SOX2干性标志蛋白在CSCs中的表达;miR-520f-3p在CSCs中的表达;经脂质体分别转染对照NC和miR-520f-3p inhibitor,并分为NC组和miR-520f-3p inhibitor组,MTS法检测各组CSCs增殖能力,Transwell实验检测各组CSCs转移能力。采用Targetscan 7.1预测软件预测miR-520f-3p的靶基因。qRT-PCR法检测miR-520f-3p对CSCs中癌症亮氨酸拉链下调因子1(LDOC1)mRNA表达的影响。结果qRT-PCR结果显示,miR-520f-3p在胶质瘤组织中的表达高于正常脑组织,其高表达与肿瘤直径及WHO分级有关(P均<0.05)。OCT4、Nanog、SOX2干细胞标志物在CSCs中表达显著上调;miR-520f-3p在CSCs中的表达高于胶质瘤细胞U87(P<0.05);转染miR-520f-3p inhibitor后,CSCs细胞增殖和转移能力均下降(P均<0.05)。TargetScan7.1预测软件和双荧光素酶报告基因试验证实LDOC1为miR-520f-3p的靶基因,miR-520f-3p inhibitor组LDOC1 mRNA表达增加(P<0.05)。结论miR-520f-3p在胶质瘤组织和CSCs中均呈高表达,且高表达与肿瘤直径及WHO分级有关;miR-520f-3p可能通过负调控LDOC1增加CSCs的干性。Objective To investigate the expression and clinical significance of miR-520f-3p in glioma tissues and glioma cancer stem cells(CSCs).Methods The expression of miR-520f-3p in 40 glioma tissues and 20 brain tissues was detected by qRT-PCR.The relationship between miR-520f-3p expression and clinical pathological characteristics in the glioma tissues was analyzed.Magnetic beads were used to sort CSCs in the glioma cell line U87.Western blotting was used to detect the expression of OCT4,Nanog,SOX2 stem marker protein in CSCs,and the expression of miR-520f-3p in CSCs.The control NC and miR-520f-3p inhibitor were transfected into CSCs with liposomes,and they were divided into the NC group and miR520f-3p inhibitor group.MTS was used to detect the proliferation ability and Transwell test was used to detect the metastasis ability of CSCs in each group.Targetscan 7.1 prediction software and dual luciferase reporter gene assay were used to study the targeting effect of miR-520f-3p on LDOC1 gene,and qRT-PCR was used to detect the effect of miR-520f-3p on LDOC1 mRNA expression in CSCs.Results The expression of miR-520f-3p in the glioma tissues was significantly higher than that in the normal brain tissues,and the high expression was significantly correlated with tumor diameter and higher WHO classification(P<0.05).The expression of OCT4,Nanog,and SOX2 stem cell markers was significantly up-regulated in CSCs;the expression of miR-520f-3p in CSCs was significantly higher than that in the glioma cells U87(P<0.05);after transfecting miR-520f-3p inhibitor,the proliferation and metastasis of CSCs decreased(both P<0.05).TargetScan 7.1 prediction software and dual luciferase reporter gene assay confirmed that LDOC1 was the target gene of miR-520f-3p,and the expression of LDOC1 mRNA in the miR-520f-3p inhibitor group increased(P<0.05).Conclusions The miR-520f-3p is highly expressed in glioma tissues and CSCs,and the high expression is associated with large tumor diameter and higher WHO classification.The miR-520f-3p may increase the s
