摘 要:目的研究参麝活络丸对脑缺血再灌注损伤大鼠脑神经元凋亡的影响。方法60只SPF级SD大鼠按体重随机分为假手术对照、模型组、参麝活络丸低剂量组、参麝活络丸中剂量组、参麝活络丸高剂量组、尼莫地平片组,每组10只。采用Zea-Longa线栓法造成脑中动脉阻塞-再灌注损伤模型。各给药组给药体积为10 ml/kg,假手术组和模型组灌胃等体积蒸馏水,连续给药7 d,观察参麝活络丸对大鼠脑缺血模型神经行为损伤评分、大脑前额叶神经元损伤、大鼠脑前额叶凋亡因子表达的影响。结果参麝活络丸各剂量组比模型对照组神经行为损伤评分显著性降低(P<0.05);参麝活络丸组比模型对照组大鼠脑前额叶神经元cyt-c、Apaf1、Caspase-9和Caspase-3表达显著性降低(P<0.05)。结论参麝活络丸可通过抑制cyt-c、Apaf1、Caspase-9、Caspase-3凋亡通路对脑中动脉阻塞-再灌注损伤模型产生治疗作用,为参麝活络丸在临床上治疗脑缺血再灌注损伤提供了试验依据。Objective To investigate the effects of Shenshehuoluo pills on neuronal apoptosis in rats with cerebral ischemia-reperfusion injury.Methods A total of 60 SPF SD rats were randomly divided into sham-operation group,model group,Shenshehuoluo high-dose group,middle-dose,low-dose group and nimodipine group,with 10 rats in each group.The animal models with middle cerebral artery occlusion-reperfusion injury were established by Zea-Longa suture method.The medication dosage was 10ml/kg by gastrointestinal administration,and the rats in sham-operation group and model group were given equal-volume distilled water for 7 days.The neurological behavioral damage scores of the cerebral ischemia model,neuronal damage in brain prefrontal lobe and expressions of apoptotic factors in prefrontal cortex were observed.Results The neurobehavioral injury scores in the different doses of Shenshehuoluo groups were significantly lower than those in model control group(P<0.05).The expression levels of cyt-c,Apaf1,Caspase-9 and Caspase-3 in the prefrontal neurons of the rats in the three Shenshehuoluo groups were significantly lower than those in model control(P<0.05).Conclusion Shenshehuoluo pills can treat the middle cerebral artery occlusion-reperfusion injury in rats by inhibiting the cyt-c,Apaf1,Caspase-9 and Caspase-3 apoptosis pathways,the experimental results provide theoretical basis for Shenshehuoluo pills in treatment of cerebral ischemia-reperfusion injury in clinical peactice.
