摘 要:目的检测心肌缺血大鼠血清和心肌组织中三磷酸腺苷(ATP)酶活性及降钙素基因相关肽(CGRP)表达,探讨大蒜素对缺血性心脏病的保护作用机制及意义。方法对成年雄性Wistar大鼠采用腹腔注射异丙肾上腺素的方法构建心肌缺血模型,随机分为模型组及大蒜素高剂量组、大蒜素中剂量组、大蒜素低剂量组,每组12只;选择12只正常大鼠作为空白对照组。对高剂量组、中剂量组、低剂量组分别用56.25、112.5、225 mg/(kg·d)的大蒜素灌胃;模型组用等容量豆油灌胃。灌胃30 d时,摘眼球取血,采用酶联免疫吸附测定法(ELISA)测定血清ATP、CGRP水平;称量各组体质量,取出心脏并称量,计算心脏指数;HE染色法观察心脏组织形态;取左心室组织匀浆,采用ELISA法检测心肌组织ATP、CGRP水平。结果模型组心脏指数高于空白组,高剂量组、中剂量组、低剂量组心脏指数均高于模型组(P均<0.05)。高剂量组、中剂量组、低剂量组心脏指数随大蒜素剂量的增加呈降低趋势(r=-0.357,P=0.026)。模型组心肌组织ATP酶活性、CGRP含量均低于空白组(P均<0.05);各剂量组心肌组织ATP酶活性均高于模型组(P均<0.05)。高剂量组、中剂量组心肌组织CGRP含量均高于模型组(P均<0.05),且心肌组织CGRP含量与大蒜素的剂量呈依赖性(r=0.342,P=0.031)。心肌组织ATP含量与CGRP呈正相关(r=0.246,P=0.029)。结论大蒜素对心肌缺血具有一定的保护作用,其机制可能与促进心肌组织ATP活性及CGRP含量有关。Objective To detect the adenosine triphosphate(ATP)activity and the calcitonin gene-related peptide(CGRP)expression in the serum and myocardial tissues of rats with myocardial ischemia,and to investigate the protective effect of allicin on ischemic heart disease rats and its mechanism and significance.Methods The myocardial ischemia models were established by intraperitoneal injection of isoproterenol in adult male Wistar rats.The rats in the model group were randomly divided into the model group and high-dose allicin,medium-dose allicin and low-dose allicin groups,with 12 in each;12 normal rats were selected as the blank control group.The rats in the high-dose group,the medium-dose group and the low-dose group were administered 56.25,112.5,and 225 mg/(kg·d)allicin,respectively;the rats in the model group and the blank group were intragastrically administered the same volume of soybean oil.After 30 days of gavage,the eyeballs were taken for blood collection.Serum ATP and CGRP levels were determined by enzyme-linked immunosorbent assay(ELISA).The mass of each group was weighed,the heart was taken out and weighed,and the cardiac index was calculated.The heart histomorphology was observed by HE staining;left ventricular tissue homogenate was taken,and ATP and CGRP levels in the myocardial tissues were detected by ELISA.Results The heart index of the model group was higher than that of the blank group,and those of the high-dose group,the medium-dose group and the low-dose group were higher than that of the model group(all P<0.05).The cardiac index of the high-dose group,medium-dose group and low-dose group decreased with the increase of allicin dose(r=-0.357,P=0.026).The activity of ATPase and CGRP in the model group were lower than those in the blank group(both P<0.05),and the activity of ATPase in each allicin dose group was higher than that in the model group(P<0.05).The content of CGRP in high-dose group and medium-dose group was higher than that in the model group(P<0.05),and the content of CGRP in the myocardi
