摘  要:目的观察不同剂量和不同给药时长龙井茶多酚提取物对小鼠肝Cyp450酶的影响。方法取40只小鼠,随机分为空白组、75 mg/kg组、150 mg/kg组、300 mg/kg组各10只,龙井茶多酚提取物组给予75、150、300 mg/(kg·d)灌胃,空白组给予超纯水0.1 mL/(10 g·BW)灌胃,各组连续灌胃7 d。取另外40只小鼠,随机分为对照组、7 d组、14 d组、28 d组各10只,7 d组、14 d组、28 d组以龙井茶多酚提取物150 mg/(kg·d)分别灌胃7、14、28 d,对照组给予超纯水0.1 mL/(10 g·BW)灌胃28 d。采用real-time PCR法检测各组肝脏Cyp3a11、Cyp 1a2、Cyp2c37、Cyp2d22 mRNA表达,采用Western blotting法检测各组肝脏Cyp3a11、Cyp1a2、Cyp2c37、Cyp2d22蛋白表达。结果相对于空白组,不同剂量的龙井茶多酚提取物各组Cyp3a11、Cyp2d22、Cyp2c37 mRNA及蛋白表达均有不同程度降低,Cyp1a2 mRNA及蛋白表达均增高(P均<0.05)。相对于对照组,不同给药时长的龙井茶多酚提取物对Cyp3a11和Cyp2d22 mRNA及蛋白表达先降低、再增高,Cyp1a2 mRNA及蛋白表达增高,对Cyp2c37 mRNA及蛋白表达降低(P均<0.05)。结论龙井茶多酚能够显著影响小鼠肝Cyp450s酶多种亚型的表达,且剂量和给药时长对Cyp450底物药物的代谢过程产生不同的影响。Objective To study the effect of Longjing tea polyphenol extracts on hepatic Cyp450 enzyme in mice.Methods Forty mice were randomly divided into three groups:the blank group,75 mg/kg,150 mg/kg and 300 mg/kg Longjing tea polyphenol extracts groups.Mice in the Longjing tea polyphenol extracts groups were given 75,150,and 300 mg/(kg·d)Longjing tea polyphenol extracts by gavage for 7 days,and the mice in the blank control group with 0.1 mL/(10 g·BW)ultrapure water by gavage for 7 days as well.Another 40 mice were randomly divided into the control group,7 d group,14 d group and 28 d group,with 10 in each.The mice in the 7 d,14 d and 28 d groups were given 150 mg/kg·d Longjing tea polyphenol extracts by gavage for 7,14,and 28 days,respectively;the mice in the control group were given 0.1 mL/(10 g·BW)ultrapure water for 28 days.The expression of Cyp3a11,Cyp1a2,Cyp2c37,and Cyp2d22 mRNA was detected by real-time PCR,and the expression of Cyp3a11,Cyp1a2,Cyp2c37,and Cyp2d22 protein was detected by Western blotting.Results Compared with the blank group,the expression levels of Cyp3a11,Cyp2d22,Cyp2c37 mRNA and protein decreased,and the expression of Cyp1a2 mRNA and protein increased in the Longjing tea polyphenol extracts groups(all P<0.05).Compared with the control group,the expression levels of Cyp3a11,Cyp2d22 mRNA and protein first decreased,and then increased,the expression of Cyp1a2 mRNA and protein increased,and the expression of Cyp2c37 mRNA and protein decreased in the Longjing tea polyphenol extracts groups(all P<0.05).Conclusions Longjing tea polyphenol could significantly affect the expression of various subtypes of Cyp450 enzyme in mice liver and its dose and duration of administration have different effects on the metabolic process of Cyp450 substrate drugs.
