摘  要:目的探索β-catenin调控NF-κB通路对大鼠前腭缝牵张成骨的作用,研究通过使用β-catenin激动剂(SB-415286)对NF-κB及相关成骨因子表达的影响。方法将45只SD大鼠随机分为三组,分别为对照组、扩弓组、SB组(扩弓时局部注射用药),对照组、扩弓组均注射赋形剂DMSO,每日1次。各组大鼠分别于扩弓1、4、7 d后处死,使用Micro-CT扫描并测量前腭缝宽度,采用HE、Masson染色观察组织形态学改变,IHC观察相关因子的变化。结果实验期间扩弓组、SB组前腭缝宽度均显著大于对照组,SB组大鼠NF-κB p65表达相对弱于扩弓组大鼠,β-catenin表达强于扩弓组大鼠(P<0.05);而SB组大鼠前腭缝成骨因子骨钙素(OCN)、骨形态发生蛋白(BMP-2)的IOD值高于扩弓组(P<0.05)。结论使用β-catenin激动剂能够下调NF-κB p65,抑制了NF-κB通路,促进BMP-2、OCN表达,加速了上颌骨前腭缝新骨的形成与钙化沉积。Objective To explore the effect of β-catenin regulating NF-κB pathway on distraction osteogenesis of premaxillary suture in rats, and to study the effect of β-catenin agonist on NF-κB and related osteogenic factors. Methods 45 SD rats were randomly divided into three groups: control group, maxillary expansion group, SB group(local injection at the time of maxillary expansion), control group and maxillary expansion group, all of which were injected with DMSO once a day. The rats in each group were killed on the 1 st, 4 th and 7 th day after the expansion. The width of the premaxillary suture was measured by Micro-CT, and the histological changes were observed by HE, Masson staining. IHC was used to observe the changes of related factors. Results During the experiment, the width of premaxillary suture in SB group was significantly wider than that in control group. The expression of p65 in SB group was lower than that in maxillary expansion group, the expression of β-catenin was stronger than that in maxillary expansion group, while the IOD value of osteocalatine(OCN), bone morphogenetic protein(BMP-2) in SB group was higher than that in maxillary expansion group(P<0.05). Conclusion β-catenin can down-regulate p65, and inhibit NF-kappa B pathway, and promote BMP-2, OCN expression, and accelerate the formation and calcification of new bone in premaxillary suture.
