摘 要:目的研究二烯丙基二硫(diallyl disulfide,DADS)是否通过下调TGF-β1/Rac1通路抑制人胃癌MGC803细胞β-catenin表达。方法实验组分为6组,对照组(Control),DADS处理组,TGF-β1处理组,TGF-β1+SB431542组,TGF-β1+DADS组,TGF-β1+NSC23766组。RT-PCR、Western blot分别检测TGF-β1、Rac1、β-catenin、E-cadherin和vimentin mRNA和蛋白水平。结果DADS下调TGF-β1、Rac1、β-catenin和vimentin,上调E-cadherin表达。TGF-β1受体阻断剂SB431542和Rac1抑制剂NSC23766下调β-catenin和vi-mentin,上调E-cadherin表达。结论DADS通过下调TGF-β1/Rac1通路抑制人胃癌MGC803细胞β-catenin表达。Aim To investigate whether diallyl disul-fide(DADS)inhibitsβ-catenin expression by down-regulating the TGF-β1/Rac1 pathway in human gastric cancer MGC803 cells.Methods The experimental group was divided into six groups,control,DADS,TGF-β1,TGF-β1+SB431542,TGF-β1+DADS,and TGF-β1+NSC23766 group.RT-PCR and Western blot were performed to detect mRNA and protein lev-els,respectively.Results DADS down-regulated TGF-β1,Rac1,β-catenin and vimentin,and up-regu-lated E-cadherin expression.TGF-β1 receptor blocker SB431542 and Rac1 inhibitor NSC23766 down-regula-tedβ-catenin and vimentin,up-regulated E-cadherin expression.Conclusions DADS inhibitsβ-catenin ex-pression by down-regulating the TGF-β1/Rac1 pathway in human gastric cancer MGC803 cells.
