摘　　要：[目的]探讨long non-coding RNA-HEIH/enhancer of zeste homolog 2 (lnc RNA-HEIH/EZH2)通路在肝癌中的作用及血清学表达中的意义。[方法]选择确诊为肝癌的早期患者30例、晚期患者30例以及健康志愿者30例,收集其临床资料及血清学样本。运用qPCR方法,检测其血清中的lncRNA-HEIH基因表达量。利用脂质体包裹lncRNA-HEIH反义寡核苷酸片段转染肝癌细胞,构建lncRNA-HEIH基因沉默的BEL-7402肝癌细胞模型,运用qPCR方法检测lncRNA-HEIH基因及EZH2基因在lncRNA-HEIH基因沉默后的表达。运用Western Blotting对肝癌细胞中EZH2蛋白进行半定量检测。运用CCK-8法对lncRNA-HEIH基因沉默前后肝癌细胞的增殖活性进行检测。[结果]早期和晚期肝癌患者血清中lncRNA-HEIH基因及EZH2基因的表达量均高于健康人,差异有统计学意义(P<0.05)。当lncRNA-HEIH基因沉默后,EZH2基因表达下降,肝癌细胞的增殖活性明显降低。[结论] LncRNA-HEIH基因可作为肝癌发生发展的潜在的血清学生物标志物。[Objective] To investigate the expression of lncRNA-HEIH gene in liver cancer patients and its relation with tumor cell proliferation. [Methods] Thirty patients with early liver cancer,30 patients with advanced liver cancer and 30 healthy subjects were enrolled in the study.The expression of lncRNA-HEIH gene in serum was detected by qPCR. Liposome-coated lncRNA-HEIH antisense oligonucleotide fragments were transfected into liver cancer BEL-7402 cells to silence lncRNA-HEIH gene,then the expressions of lncRNA-HEIH gene and EZH2 gene were detected. EZH2 protein expression in BEL-7402 cells was detected by Western blot. The proliferation activity of BEL-7402 cells was detected by CCK-8 before and after lncRNA-HEIH gene silencing. [Results] The expression of lncRNA-HEIH gene and EZH2 gene in serum of patients with early and advanced liver cancer was higher than that of healthy subjects(P <0.05). When lncRNA-HEIH gene was silenced,the expression of EZH2 gene decreased,and the proliferation activity of liver cancer cells inhibited significantly.[Conclusion] The expression of lncRNA-HEIH gene is upregulated in liver cancer,which might be used as a potential serological biomarker for liver cancer.
