摘  要:基于RAPD分子标记技术,通过计算遗传相似系数、UPGMA聚类、主成分分析,研究了10株供试杏鲍菇菌株的遗传多样性。利用筛选出的8条谱带清晰、多态性较好的引物合计扩增出47个位点,其中36个多态性位点,多态性比率为77%,遗传相似系数在0.556~1.000。结果表明,供试菌株在0.62水平上被聚为1类,在0.68水平上可分为3个类群。RAPD分析结果与主成分分析结果一致,说明RAPD技术能够作为快速鉴别杏鲍菇菌株亲缘关系的一种方法,有利于对杏鲍菇种质资源管理、优良品种选育等。Based on RAPD technology, the genetic diversity of 10 Pleurotus eryngii strains was studied by calculating genetic identity, UPGMA clustering and PCoA. A total of 47 polymorphic bands, among which 36 polymorphic bands were amplified by 8 selected primers which have clear and polymorphic richness strips, and the percentage of polymorphic loci was 77%, the genetic identity was 0.556~1.000. The tested strains could be clustered into one group at the genetic similarity level of 0.62, and clustered into 3 groups at the level of 0.68. The results of UPGMA analysis were accordance with PCoA, which showed that RAPD could be effective to distinguish the species among P. eryngii. These results could provide valuable references for management of P. eryngii germplasm resources and breeding research.
