摘 要:目的 检测宫颈癌患者肿瘤组织miR-34a、miR-145表达及人乳头瘤病毒(HPV)阳性表达率,并分析其相关性。方法 选取2016年9月-2018年9月浙江省台州医院收治的疑似宫颈癌患者120例为研究对象,其中宫颈癌患者46例,宫颈上皮内瘤变(CIN)患者Ⅱ~Ⅲ级48例,宫颈炎患者26例。利用第二代杂交捕获法对HPV-DNA进行分型检测,利用HPV E6/E7mRNA检测试剂盒检测组织细胞中HPV E6/E7mRNA表达情况,利用实时定量PCR(qRT-PCR)法检测宫颈组织中miR-34a、miR-145水平。结果 宫颈癌组患者宫颈组织中miR-34a、miR-145表达水平低于宫颈炎组、CINⅡ~Ⅲ组患者,CINⅡ~Ⅲ组患者宫颈组织中miR-34a、miR-145表达水平低于宫颈炎组患者,差异均有统计学意义(P<0.05)。宫颈癌组患者肿瘤组织中HPV-DNA阳性表达率、HPV E6/E7 mRNA高载量率显著高于宫颈炎组、CINⅡ~Ⅲ组患者,CINⅡ~Ⅲ组患者组织中HPVDNA阳性表达率、HPV E6/E7 mRNA高载量率显著高于宫颈炎组患者,差异均有统计学意义(P<0.05)。宫颈癌患者肿瘤组织中miR-34a、miR-145表达与HPV感染、HPV E6/E7 mRNA表达呈负相关(P<0.01)。结论 宫颈癌患者肿瘤组织中miR-34a、miR-145低表达,与HPV感染负相关。Objective To detect the expressions of miR-34 a and miR-145 in cervical cancer tissues and the positive expression rate of human papillomavirus(HPV),analyze the correlations.Methods A total of 120 patients suspected of cervical cancer treated in Taizhou Hospital of Zhejiang Province from September 2016 to September 2018 were selected as study subject,including 46 patients with cervical cancer,48 patients with cervical intraepithelial neoplasia(CIN)Ⅱ-Ⅲand 26 patients with cervicitis.HCⅡwas used to detect HPV-DNA.HPV E6/E7 mRNA detection kit was used to detect HPV E6/E7 mRNA expression.qRT-PCR was used to detect the levels of miR-34 a and miR-145 in cervical tissues.Results The expression levels of miR-34 a and miR-145 in cervical cancer group were statistically significantly lower than those in cervicitis group and CINⅡ-Ⅲgroup,the expression levels of miR-34 a and miR-145 in CINⅡ-Ⅲgroup were statistically significantly lower than those in cervicitis group(P<0.05).The positive expression level of HPV-DNA and high load rate of HPV E6/E7 mRNA in cervical cancer group were statistically significantly higher than those in cervicitis group and CINⅡ-Ⅲgroup,the positive expression level of HPV-DNA and high load rate of HPV E6/E7 mRNA in CINⅡ-Ⅲgroup were statistically significantly higher than those in cervicitis group(P<0.05).The expressions of miR-34 a and miR-145 in cervical cancer group were negatively correlated with HPV infection and HPV E6/E7 mRNA expression(P<0.01).Conclusion The expressions of miR-34 a and miR-145 are low in cervical cancer tissues,which are negatively correlated with HPV infection.
