摘　　要：目的:去卵巢和睡眠剥夺均可对雌性小鼠的焦虑行为产生影响,然而睡眠剥夺是否会加重去卵巢小鼠的焦獉獉虑情绪及机制尚不清楚。因此,本研究主要探讨去卵巢或联合睡眠剥夺对雌性小鼠焦虑样行为的影响及柴胡加龙骨牡蛎汤有效部位(CHJLMD70E)的干预作用。方法:去卵巢和去卵巢合并跑步机24 h睡眠剥夺小鼠灌胃给予獉獉CHJLMD70E(48 mg·kg-1,连续7 d),然后进行高架十字迷宫测试,促肾上腺皮质激素(Adrenocorticotropic Hormone,ACTH)、皮质酮(Corticosterone,CORT)及褪黑素(Melatonin,MT)含量的测定。跑步机睡眠剥夺组小鼠于给药第6 d置于跑步机中(3 s运转,12 s停止,速度为1 m·min-1)。结果:去卵巢组和去卵巢合并跑步机睡眠獉獉剥夺组小鼠的开臂次数比例和开臂时间比例均减少,MT含量减少,CORT含量增加;CHJLMD70E可拮抗上述变化。结论:CHJLMD70E可能是通过增加MT水平,调节下丘脑-垂体-肾上腺轴(Hypothalamic-Pituitary-Adrenal,獉獉HPA),改善去卵巢和去卵巢合并睡眠剥夺小鼠的焦虑样行为。Objective: Ovariectomy and sleep deprivation can affect the anxiety behavior of female mice,but whether sleep deprivation can aggravate the anxiety of ovariectomized mice and its mechanism are not clear. Therefore,the present study was performed to determine the interventive effects of the active component of Chaihu Jia Longgu Muli Decoction( CHJLMD70 E) on anxiety-like behavior in ovariectomized alone or in a combination with sleep deprived( SD) female mice.Methods: CHJLMD70 E ( 48 mg · kg-1·day-1,7 consecutive days,i. g.) was given to the ovariectomized mice or in a combination with SD for 24 h using treadmill. Anxiety-like state was tested in the elevated plus maze.Moreover,adrenocorticotropic hormone( ACTH),corticosterone( CORT) and melatonin( MT) levels in the serum were measured. On the 6 th after the beginning of administration,the SD mice ran on treadmill for 24 h( 3 s treadmill on followed by 12 s off,1 m/min).Results: In ovariectomized group and ovariectomized combined with treadmill sleep deprivation group,the percentage of open-arm time and open-arm entries decreased,and the content of MT decreased and the content of CORT increased.CHJLMD70 E could antagonize the above changes.Conclusion: Our findings suggest that CHJLMD70 E may improve the increased anxiety-like behavior in ovariectomized alone or in a combination with SD female mice by increasing level of serum MT and regulating HPA axis function.
