摘 要:目的探讨利用1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)进行腹腔注射建立帕金森病(PD)大鼠模型,评价其血液学及临床病理改变。方法选取爬杆得分为0分的雄性SD大鼠16只,随机分为对照组(6只)和MPTP模型组(10只)。对MPTP模型组大鼠连续7d腹腔注射MPTP(30mg/kg),建立PD大鼠模型。利用血常规与血生化评价大鼠血液学。观察各组大鼠行为学(旷场试验、爬杆试验)的变化,用HE染色法观察黑质区病理变化及免疫组化法测定酪氨酸羟化酶(TH)阳性神经元数量。结果与对照组相比,MPTP模型组大鼠在旷场试验中的方格间穿行次数和站立次数明显减少(P<0.05),爬杆试验得分明显增加(P<0.05)。MPTP模型组大鼠的血生化指标直接胆红素明显降低(P<0.05),其血常规指标中血红蛋白、红细胞、白细胞、中性粒细胞、淋巴细胞、单核细胞数均明显或极明显高于对照组(P<0.05,P<0.01),其他指标差异均无统计学意义(P>0.05)。与对照组相比,MPTP模型组大鼠的黑质区神经元细胞皱缩、体积减小、神经元数量减少、细胞周围间隙增宽,TH+神经元数目明显降低。结论利用MPTP诱导的大鼠出现明确的PD行为学特征和病理损伤。PD大鼠的肝功、肾功、血糖、血脂等指标无明显影响,而其血常规指标均明显增高,提示出现炎症。Objective To establish the model rats with Parkinson’s Disease(PD)by the intraperitoneal injection with 1-methyl-4-phenvl-1,2,3,6-tetrahyvdropvridine(MPTP)and evaluate the hematological and clinicopathological changes.Methods A total of 16 male SD rats with the score of zero in the pole test were selected and randomly divided into the control group with 6 rats and the MPTP model group with 10 rats.The rats in the MPTP model group were intraperitoneally injected with MPTP at the dosage of 30 mg/kg for 7 consecutive days to establish the PD rat model.The blood routine and blood biochemistry were used to evaluate rats’hematology.The changes of rats’behaviors in the open-field test and pole test were observed in each group.The pathological changes of substantia nigra were observed by HE staining and the number of Tyrosine hydroxylase(TH)positive neurons were determined by immunohistochemistry.Results Compared with the control group,the times of crossing and rearing in the open field test were significantly reduced in the MPTP model group(P<0.05),and the score of the pole test was significantly increased(P<0.05).The direct bilirubin of the blood biochemical indexes in the MPTP model group was significantly decreased(P<0.05)and the numbers of the blood routine indexes including hemoglobin,erythrocyte,white blood cell,neutrophil,lymphocyte and monocyte in the MPTP model group were significantly or extremely significantly higher than those in the control group(P<0.05 or P<0.01).There were no significant differences in the other indexes(P>0.05).Compared with the control group,the neurons of the substantia nigra shrank and decreased in volume,the number of neurons decreased,the pericellular space widened,and the TH+neurons decreased significantly in the MPTP model group.Conclusion The MPTP-induced rats showed definite PD behaviors and pathological damages.There were no significant effect on the indexes including liver function,kidney function,blood sugar and blood lipid in the PD rats,while the blood routine indexes
