摘  要:目的探讨银杏叶提取物对血管性痴呆大鼠记忆力及γ-氨基丁酸转运体-1(GAT-1)、cAMP反应原件结合蛋白(CREB)表达的影响。方法 SPF级成年雄性SD大鼠(30只)都建立了血管性痴呆模型,然后随机分为三组-对照组、奥拉西坦组与银杏组,每组各10只大鼠。对照组给予生理盐水治疗,奥拉西坦组给予奥拉西坦治疗,银杏组给予银杏叶提取物治疗,都持续用药14d,检测大鼠记忆力与GAT1、CREB表达变化情况。结果三组造模后1d的逃避潜伏期对比差异无统计学意义(P>0.05),银杏组与奥拉西坦组造模后7d、14d的逃避潜伏期低于对照组,银杏组低于奥拉西坦组,对比差异都有统计学意义(P<0.05)。银杏组与奥拉西坦组造模后14d的颈动脉微血管密度与海马组织的GAT1、CREB蛋白相对表达水平高于对照组,银杏组高于奥拉西坦组,对比差异都有统计学意义(P<0.05)。银杏组与奥拉西坦组造模后14d海马组织的白介素(IL)-1β、IL-6低于对照组,银杏组低于奥拉西坦组,对比差异都有统计学意义(P<0.05)。结论银杏叶提取物在血管性痴呆大鼠中的应用可促进海马组织GAT1、CREB表达,提高颈动脉微血管密度,抑制炎症因子的释放,从而改善大鼠记忆力。Objective To investigate the effects of ginkgo biloba extract on memory and expression ofγ-aminobutyric acid transporter-1(GAT-1)and cAMP response element binding protein(CREB)in rats with vascular dementia.Methods SPF adult male SD rats(30 rats)were established with vascular dementia model,and then were equally randomly divided into three groups-control group,oxiracetam group and ginkgo biloba group,with 10 rats in each group.The control group were treated with normal saline,the oxiracetam group were treated with oxiracetam,and the ginkgo group were treated with ginkgo biloba extract for 14 days.The changes in memory and expression of GAT1 and CREB in rats were detected.Results There were no significant difference in escape latency compared among the three groups at 1 dafter modeling(P>0.05),the escape latency of the ginkgo group and the oxiracetam group were lower than those of the control group at 7 dand 14 dafter modeling,and the ginkgo group were lower than that of the control group,there were also statistically significant differences compared between the ginkgo group and the oxiracetam group(P<0.05).The carotid microvessel density and the relative expression levels of GAT1 and CREB proteins in hippocampal tissues were higher in the ginkgo group and the oxiracetam group at 14 days after modeling,and the ginkgo group were higher than the oxiracetam group.(P<0.05).In the ginkgo group and the oxiracetam group,the levels of interleukin(IL)-1βand IL-6 in the hippocampus were lower than those in the control group and the ginkgo group were lower than that of the oxiracetam group at 14 days after modeling.0.05).Conclusion The application of Ginkgo biloba extract in vascular dementia rats can promote the expression of GAT1 and CREB in hippocampus,increase the density of carotid microvessels,inhibit the release of inflammatory factors,and improve memory in rats.
