摘  要:副溶血性弧菌是一种革兰氏阴性嗜盐致病菌,被认为是多血清型引起的主要食源性致病菌.通过全基因组测序技术,实现对K3、K29、K61菌株的测序.并且使用相关软件对测序数据进行基因预测和功能注释从而实现抗原决定簇的破译.鉴定的3个血清型的K抗原基因簇,表现出高程度的遗传多样性.根据K抗原基因决定簇的特异基因,建立了用于检测和鉴定这3种K血清型的液相芯片检测体系.本研究中破译的副溶血弧菌K抗原基因决定簇以及基于分子技术建立的检测分型体系,为高效的细菌检测提供了可能.Vibrio parahaemolyticus is a Gram-negative halophilic pathogen and is considered to be a major food-borne pathogen caused by multiple serotypes. Sequencing of K3, K29, and K61 strains was achieved by whole genome sequencing technology. Using relevant software to perform genetic prediction and functional annotation on the sequencing data to decipher antigenic determinant. Three serotypes of the K antigen gene cluster were identified and found to exhibit a high degree of genetic diversity. Based on the specific genes of the K antigen gene determinant, a suspension array detection system for detecting and identifying the three K serotypes was established. The decomposed V. parahaemolyticus K antigen gene determinant and the detection and typing system based on molecular technology in this study provide the possibility of efficient bacterial detection.
