摘  要:目的观察卵巢癌组织微小RNA-204(miR-204)、脑源性神经营养因子(BDNF)表达变化,并探讨其临床意义。方法选择卵巢癌患者89例、因子宫良性病变行子宫及附件切除手术且经术后组织病理检查证实卵巢正常患者20例,取手术切除的卵巢癌组织和正常卵巢组织,采用RT-qPCR法检测miR-204、BDNF表达,比较卵巢癌组织与正常卵巢组织miR-204、BDNF表达差异。分析卵巢癌组织miR-204、BDNF表达与患者临床病理特征和预后的关系。结果卵巢癌组织miR-204相对表达量低于正常卵巢组织,BDNF相对表达量高于正常卵巢组织(t分别为38.904、11.799,P均<0.01)。Pearson线性相关分析显示,卵巢癌组织miR-204相对表达量与BDNF相对表达量呈负相关关系(r=-0.603,P<0.01)。卵巢癌组织miR-204、BDNF相对表达量均与组织分化程度、临床分期有关(P均<0.05),与年龄、病理类型、淋巴结或腹膜转移、血清CA125水平及腹水量无关(P均>0.05)。以卵巢癌组织miR-204、BDNF相对表达量的均数为临界值,将患者分为miR-204高表达者44例与低表达者45例、BDNF高表达者43例与低表达者46例。miR-204低表达者3年总体生存率低于miR-204高表达者(χ^2=3.901,P<0.05),BDNF高表达者3年总体生存率低于BDNF低表达者(χ^2=4.690,P<0.05)。结论卵巢癌组织miR-204低表达、BDNF高表达,二者表达变化与临床分期、组织分化程度和患者预后有关。miR-204、BDNF有可能成为卵巢癌分子诊断、靶向治疗和预后评估的生物标志物。Objective To investigate the expression changes of microRNA-204(miR-204)and brain-derived neurotrophic factor(BDNF)in the ovarian cancer tissues and to explore its clinical significance.Methods Eighty-nine cases of ovarian cancer tissues and 20 cases of normal ovarian tissues confirmed by histopathological examination were selected.The ovarian cancer tissues and normal ovarian tissues were removed.The expression levels of miR-204 and BDNF were detected by RT-qPCR.The expression difference of miR-204 and BDNF between ovarian cancer and normal ovarian tissues were compared.The relationships of the expression of miR-204 and BDNF in the ovarian cancer tissues with the clinicopathological characteristics and prognosis were analyzed.Results The relative expression of miR-204 in the ovarian cancer tissues was lower than that in the normal ovarian tissues,and the relative expression of BDNF was higher than that in the normal ovarian tissues(t=38.904,11.799,both P<0.01).Pearson linear correlation analysis showed that there was a negative correlation between the relative expression of miR-204 and BDNF in the ovarian cancer tissues(r=-0.603,P<0.01).The relative expression of miR-204 and BDNF in the ovarian cancer tissues was related to the degree of tissue differentiation and TNM stage(both P<0.05),but not to age,pathological type,lymph node or peritoneal metastasis,serum CA125 level or ascites volume(all P>0.05).According to the mean value of miR-204 and BDNF relative expression in the ovarian cancer tissues,the patients were divided into 44 cases with high expression of miR-204,45 cases with low expression,43 cases with high expression of BDNF,and 46 cases with low expression.The 3-year overall survival rate of patients with low expression of miR-204 was lower than that of patients with high expression of miR-204(χ^2=3.901,P<0.05),and the 3-year overall survival rate of patients with high expression of BDNF was lower than that of patients with low expression of BDNF(χ^2=4.690,P<0.05).Conclusion The low expression of miR
