摘 要:目的探讨皮肤鳞状细胞癌组织中的差异表达基因。方法从基因表达综合数据库(GEO)在线数据库中下载皮肤鳞状细胞癌(cSCC)组织相关的基因表达谱芯片数据集GSE42677和GSE53462,利用Limma包筛选肿瘤组织和正常皮肤组织之间的差异表达基因;利用DAVID工具对差异表达基因进行基因本体论(GO)和功能富集分析;利用STRING数据库分析差异表达基因之间的相互作用,并利用Cytoscape软件构建蛋白质之间相互作用网络,筛选核心基因和模块。结果两组芯片数据中共同上调表达基因43个,共同下调表达基因65个,这些差异基因主要富集在细胞增殖、皮肤发展、先天免疫反应、Toll样受体、趋化因子、转录调控、氨基酸代谢和p53信号通路;构建了这些基因之间的相互作用网络,获得cSCC相关前20核心基因,分别为CXCL8、CCND1、KIT、PI3、SPP1、PLAU、GATA3、PLAUR、CDKN2A、S100A7、KRT16、CXCL1、DEFB4A、ISG15、KRT2、S100A12、KRT6B、ISG20、OASL和IFI6。结论cSCC组织与正常皮肤组织之间存在大量的差异表达基因,CXCL8、CCND1、KIT、PI3、SPP1、PLAU、GATA3、PLAUR、CDKN2A和S100A7等基因及其介导的信号通路在cSCC的发生过程中可能起着重要作用。【Objective】To explore differentially expressed genes(DEGs)between cutaneous squamous cell carcinoma(CSCC)and normal skin tissues.【Methods】The CSCC related gene expression profile datasets GSE42677 and GSE53462 were download from the Gene Expression Omnibus(GEO)database.The DEGs between tumor tissues and normal skin tissues were identified by using the limma R software package.The DAVID tools were used to conduct the Gene Ontology(GO)and functional enrichment analysis of these DEGs.The interactions between DEGs were analyzed using STRING database.The protein-protein interaction(PPI)networks was constructed by Cytoscape software and they were used to obtain the core genes.【Results】There were 43 genes were up-regulated and 65 genes were down-regulated in CSCC compared with normal skin tissue.These genes were mainly enriched in cell proliferation,skin development,innate immune response,Toll-like receptor signaling pathway,chemokine signaling pathway,transcriptional regulation,amino acid metabolism,and p53 signaling pathway.The PPI network was constructed,and the top 20 core genes included CXCL8,CCND1,KIT,PI3,SPP1,PLAU,GATA3,PLAUR,CDKN2A,S100A7,KRT16,CXCL1,DEFB4A,ISG15,KRT2,S100A12,KRT6B,ISG20,OASL and IFI6,were selected from this network.【Conclusion】There are a large number of differentially expressed genes between CSCC tissues and normal skin tissues.The DEGs such as CXCL8,CCND1,KIT,PI3,SPP1,PLAU,GATA3,PLAUR,CDKN2A,S100A7 and their mediated pathways may be play an important role in the development of CSCC.
