摘 要:以凌云白毫茶嫩芽为外植体,建立凌云白毫茶无菌体系,探讨抗褐化措施,建立白毫茶组织培养体系,为白毫茶优良品种产业化种植提供种苗技术基础。结果表明:以白毫茶树带腋芽茎段和顶芽为外植体,浓度为0.1%HgCl2溶液对外植体进行灭菌10 min,污染率最低;在此基础上,预处理采用0.1%高锰酸钾溶液浸泡10 min,抗褐化效果和降低污染率都较明显,褐化率低至30%;选用腋芽茎段的褐化率和污染率均比顶芽低,在培养基中添加活性炭1.5 mg/L,能降低褐化率至16.7%。凌云白毫茶初代培养使用6-BA诱导腋芽效果好,浓度为2.0 mg/L的诱导率达到100%;对无菌芽增殖来说较适宜的培养基为MS+2.0 mg/L 6-BA+0.1 mg/L NAA+7g/L琼脂+30 g/L蔗糖,增殖倍数为3.3,长势最好。In this experiment,the buds of Lingyun Baihao tea were used as explants to establish the sterile system of Lingyun Baihao tea,and the anti-browning measures were discussed,in order to establish the tissue culture system and provide the seedling technical basis for the industrialization of Baihao tea varieties.The results showed that the stems with axillary buds and the top buds were used as explants,and the concentration of 0.1%HgCl2 solution was used to sterilize for 10 min,the contamination rate was the lowest.On this basis,0.1%potassium permanganate solution was used to soak the explants for 10 min as pretreatment,the anti-browning effect and the pollution reduction rate were obvious,the browning rate was as low as 30%.The browning rate and pollution rate of the axillary bud stems were lower than that of the top buds,and the browning rate could be reduced to 16.7%by adding 1.5mg/L active carbon in the medium.In the primary culture of Lingyun Baihao tea,6-BA was used to induce axillary buds,and the induction rate was 100%at a concentration of 2.0 mg/L.The suitable medium for sterile buds proliferation was MS+2.0mg/L 6-BA+0.1mg/L NAA+7g/L agar+30 g/L sucrose,the proliferation multiple was 3.3,and the growth potential was the best.
