摘  要:目的探讨适配子微流体芯片检测外周血循环肿瘤细胞在鼻咽癌中的诊断价值。方法选取30例未接受放化疗的鼻咽癌患者作为研究对象,治疗前后分别采集患者外周血。10例鼻咽良性疾病和20例健康体检者作为对照。样本通过适配子微流体芯片计数循环肿瘤细胞。评价适配子微流体芯片检测的重复性、敏感性、特异性。结果鼻咽癌治疗前循环肿瘤细胞的中位数是16,治疗后是6,对照组是2。检测15个临床样本,符合率100%。适配子微流体芯片检测的敏感性是77%特异性是86%。治疗前鼻咽癌患者的循环肿瘤细胞数与临床分期有相关性,治疗后的循环肿瘤细胞数与临床分期没有相关性。结论适配子微流体芯片具有较好的敏感性和特异性,外周血循环肿瘤细胞可以用于鼻咽癌的辅助诊断。Objective To explore the diagnosis value of circulating tumor cells in nasopharyngeal carcinoma patients using microfluidic chip.Methods 30 patients with nasopharyngeal carcinoma who did not receive radiotherapy or chemotherapy were enrolled in study.Peripheral blood were drew before and after the therapy.10 patients with benign nasopharyngeal diseases and 20 healthy donors were analyzed for comparison.Samples were enriched for circulating tumor cells using aptamer microfluidic chip.Circulating tumor cells were analyzed for repeatability.The sensitivity and specificity of aptamer microfluidic chip were calculated.Results The median circulating tumor cells were 16 at pre-treatment and 6 at after-treatment,2 in the control group.The repeatability was 100%in 15 patient samples.The sensitivity and specificity of aptamer microfluidic chip were 77%and 86%respectively.The number of circulating tumor cells in pre-treatment patients were relative to pathological stage,but were not in after-treatment.Conclusion The sensitivity and specificity of aptamer microfluidic chip are good and peripheral blood circulating tumor cells test can be used in the diagnosis of nasopharyngeal carcinoma.
