高效液相色谱法同时测定17种黄色系色素

摘　　要：目的:建立HPLC法同时检测(碱性黄87、碱性橙31、日落黄、酸性黄36、金胺O、金橙II、柠檬黄、碱性橙G、碱性橙22、酸性橙3、金橙Ⅰ、酸性黄1、金橙G、金橙Ⅳ、二甲基黄、亮黄、甲基橙)共17种黄色系色素的方法,并将此方法应用于中药及饮片非法添加色素的检测中。方法:选用资生堂CAPCELL PAK C18,ACR(250×4.6 mm,5μm)色谱柱,以乙腈-0.02 mol·L^-1醋酸铵水溶液(冰醋酸调至p H5.2)进行梯度洗脱,流速0.6mL·min^-1,柱温为20~22℃,在不同波长下(364 nm、410 nm、430 nm和480 nm)进行检测。结果:17种水溶性红色素在1~50μgl/mL范围内线性关系良好,精密度、稳定性RSD均小于5%,加标回收率在80.00%~120.00%之间;结论:方法操作简单、快速、准确、重现性好、灵敏度高。适用于中药材及饮片中黄色系色素的非法染色现象的筛查。The method of high performance liquid chromatography(HPLC)was established for simultaneous detecting seventeen yellow pigments(Basic Yellow 87,Basic Orange 31,Sunset Yellow,Acid Yellow 36,Auramine O,Golden Orange II,Lemon Yellow,Basic Orange G,Basic Orange 22,Acid Orange 3,Golden Orange 1,Acid Yellow 1,Golden Orange G,Golden Orange IV,Dimethyl Yellow,Bright Yellow,Helianthine),and applied to detect illegally added pigments in Chinese medicinal materials and pieces.Methods:The Shiseido CAPCELL PAK C18 column(250×4.6 mm,5μm)was used to separate the samples at 20~22℃with a step linear gradient using acetonitrile-0.02 mol·L^-1 ammonium acetate solution(acetic acid adjust to p H5.2)at a flow rate of 0.6 mL·min^-1,and detection was performed at different wavelengths(364 nm、410 nm、430 nm和480 nm).Results:Seventeen kinds of yellow pigments had good linear relationship in the range of 1~50 mg/mL,the RSD of precision and stability were less than 5%,and the recovery was between 80%and 120%;Conclusion:The method was simple,rapid,accurate,good reproducibility and high sensitivity and suitable for the screening of the illegal dyeing of yellow pigments in traditional Chinese medicine and herbals.experiment center for many years.This paper intents to further promote the overall management level of laboratory and satisfy the need of experiment teaching by means of micro-management.
