摘  要:目的研究热休克蛋白90(HSP90)及c-Myc在小鼠肠道腺瘤癌变中的表达及相关性.方法采集5周及8周ApcΔ716/+小鼠的肠道标本,通过Western blot、免疫组织化学染色及RT-PCR明确肠道腺瘤癌变组织中HSP90及c-Myc的表达情况及相关性.结果腺瘤癌变组织中HSP90及c-Myc的Western blot条带均较良性腺瘤明显增强;免疫组织化学染色见腺瘤癌变组织中HSP90及c-Myc均着色明显,其相对表达量较良性腺瘤明显升高(P均<0.001);腺瘤癌变组织中HSP90及c-Myc的mRNA的相对值较良性腺瘤明显升高(P均<0.001);HSP90及c-Myc的mRNA表达具有明显相关性(r=0.710,P=0.002).结论肠道腺瘤癌变组织中HSP90及c-Myc的表达均明显提高,二者表达具有明显的相关性,说明HSP90可能与c-Myc相互作用,在腺瘤癌变中发挥作用.Objective To investigate the expression and correlation of heat shock protein 90(HSP90)and c—Myc in the carcinogenesis of intestinal adenoma in mice.Methods The intestinal specimens of 5—week—old and 8—week—old ApcA716/+mice were collected.Western blot,immunohistochemical staining,and RT-PCR were performed to determine the expression and correlation of HSP90 and c—Myc in adenocarcinoma.Results The Western blot strips of HSP90 and c—Myc extracted from adenocarcinoma were obviously enhanced compared with benign adenomas.The adenocarcinoma cells were stained strongly,and the relative expression values of HSP90 and c—Myc in adenocarcinoma were significantly increased compared with benign adenoma(P all<0.001).The relative mRNA values of HSP90 and c—Myc in adenocarcinoma were significantly higher than those of benign adenomas(P all<0.001).The niRNA expression of HP90 and c—Myc in intestinal adenocarcinoma showed a positive correlation(R=0.710,P=0.002).Conclusion The expressions of HSP90 and c—Myc in intestinal adenocarcinoma increases significandy compared with benign adenoma,and there is positive correlation between the expression of HSP90 and c—Myc.So HSP90 may interact with c—Myc and play a role in carcinogenesis of intestinal adenoma.
