摘  要:目的分析睡眠剥夺对大鼠学习记忆和海马区p-ERK1/2蛋白表达的影响。方法将60只雄性SD大鼠随机分为对照组和睡眠剥夺组,采用多平台水环境法建立睡眠剥夺模型。采用Morris水迷宫检测对照组和睡眠剥夺组大鼠定位航行和空间搜索能力的变化,采用蛋白免疫印迹法检测海马区p-ERK1/2蛋白表达的变化。结果与对照组相比,睡眠剥夺组大鼠逃避潜伏期明显延长,穿越原平台位置的次数和平台所在象限游泳时间占总时间的百分比减少(P<0.05);蛋白免疫印迹法结果显示,睡眠剥夺组海马区p-ERK1/2表达水平低于对照组(P<0.05)。结论睡眠剥夺可损害大鼠的学习记忆,其机制可能与ERK信号通路抑制有关。Objective:To analyze the effects of sleep deprivation on learning and memory and the expression of p-ERK1/2 protein in hippocampus of rats.Methods:Sixty male SD rats were randomly divided into two groups:control group and sleep deprivation group.Morris water maze was used to observe the changes of navigation and spatial search ability of rats in the control group and sleep deprivation group.The expression of p-ERK1/2 protein in hippocampus was detected by Western blotting.Results:Compared with the control group,the escape latency of sleep in deprived rats was significantly prolonged,and the number of times of crossing the original platform position and the percentage of swimming time in the platform quadrant were decreased(P<0.05).The results of Western blotting showed that the expression of p-ERK1/2 in hippocampus of sleep deprived rats was lower than that of the control group(P<0.05).Conclusion:Sleep deprivation can damage the learning and memory of rats,which may be related to the inhibition of ERK signal pathway.
