槐耳联合化疗药对人胃癌细胞的影响

摘　　要：目的观察不同提取部位的槐耳粗提物-槐耳清膏(槐耳水提物)和槐耳醇提物联合化疗药物(顺铂和5-氟尿嘧啶)对不同人胃癌细胞(MGC-803、BGC-823、AGS、HGC-27)的影响,分析槐耳醇提物的优势以及筛选出槐耳粗提物与化疗药物联合的最佳组合,初步探讨槐耳醇提物增加化疗药物敏感性的分子机制.方法通过CCK-8实验检测不同浓度下的槐耳清膏、醇提物、顺铂(DDP),5-氟尿嘧啶(5-Fu)单独作用于胃癌细胞24h后的细胞存活率;将槐耳清膏、醇提物分别与DDP、?5-Fu联合处理胃癌细胞24h后,通过CCK-8实验比较存活率.通过Western blot实验检测槐耳醇提物作用胃癌细胞24h后,相关耐药蛋白如多药耐药关联蛋白1的差异表达情况.结果槐耳清膏、醇提物和DDP、5-Fu在单用及联合下,均对不同胃癌细胞有显著的体外抑制作用,并呈剂量依赖性.在不同胃癌细胞中相同抑制率下,醇提物的用药浓度远低于槐耳清膏,且与DDP联合对胃癌细胞的体外抑制最为强烈.槐耳醇提物作用胃癌细胞后,MRP1的表达水平下降.结论在体外实验中,槐耳醇提物的抗肿瘤增殖效用优于槐耳清膏,槐耳醇提物与顺铂联合使用可能会更有效的抑制肿瘤的发展.槐耳醇提物增加化疗药物的敏感性可能与MRP1的表达水平下降有关.Objective To observe the influences of different extracts from Huaier,Huaier Cream(Huaier aqueous extract)and Huaier n-butanol extract combined with chemotherapeutic drugs(cisplatin and 5-fluorouracil)on different human gastric cancer cells(MGC-803,BGC-823,AGS,and HGC-27),to explore the advantages of the Huaier n-butanol extract and the best combination of Huaier and chemotherapy drugs,preliminary discussion on molecular mechanism of the increase of chemotherapy drug sensitivity.Methods CCK-8 assay was used to detect gastric cancer cell viability after treatment with a series of concentrations of Huaier Cream,Huaier n-butanol extract,cisplatin(DDP)and 5-fluorouracil(5-Fu)for 24 h.The effects of combination of Huaier Cream and Huaier n-butanol extract with DDP and 5-Fu on gastric cancer cells for 24h were compared by CCK-8 assay.Western blot analysis was used to detect the differential expression of related drug resistance proteins such as multidrug resistance-associated protein 1 after GC cells were treated by Huaier n-butanol for 24h.Results The viabilities of different gastric cancer cells were significantly inhibited by Huaier Cream,Huaier n-butanol extract,DDP and 5-Fu alone or in combination in vitro,and they were in a dose-dependent manner.In addition,the concentration of Huaier n-butanol extract was much lower than Huaier Cream at the same inhibition rate in different gastric cancer cells.Besides,the inhibitory effect of combination of Huaier n-butanol extract and DDP on gastric cancer cells was the strongest in vitro.After treated by Huaier n-butanol extract,the expression level of MRP1 was decreased in GC cells.Conclusion In vitro,the anti-tumor proliferative effect of Huaier n-butanol extract is better than Huaier Cream(clinical drug:Jinke Huaier granule),and the combination of Huaier n-butanol extract and cisplatin may inhibit tumors more effectively.In addition,the sensitivity of the chemotherapeutic drug to increase by Huaier n-butanol extract may be related to the decreased expression level of MRP1.
