摘 要:目的:探讨不同浓度的凉血活血方对宫颈癌SiHa细胞增殖和凋亡的影响。方法:体外培养宫颈癌SiHa细胞,选取合适的细胞数进行铺板,将凉血活血方(浓度为1.125、2.228、3.31、4.37、5.41 mg/ml)作用于SiHa细胞24 h、48 h,MTT及CCK-8法检测细胞的增殖情况;流式细胞仪检测药物作用24 h细胞的凋亡率,并用光镜观察此时的细胞状态;蛋白免疫印迹法(Western-blot)检测药物(浓度为1.125、2.228、3.31 mg/ml)作用24 h后Bcl-2及Bax的表达情况。结果:MTT及CCK-8结果显示:药物作用24 h,除1.125 mg/ml外,其余浓度的增殖抑制率均明显高于空白对照组(P<0.05),并随药物浓度的增加而升高(P<0.05);药物作用48 h,除MTT中1.125 mg/ml外,其余各浓度的抑制率均明显高于空白对照组(P<0.05)。光镜下观察显示:除1.125 mg/ml外,其余组可见细胞的体积变小、变形,皱缩、变圆等凋亡形态。细胞凋亡结果显示:各浓度的凉血活血方均可促进SiHa细胞凋亡。Western-blot结果显示:除1.125 mg/ml外,其余浓度可使Bcl-2/Bax的比值下调。结论:一定浓度的凉血活血方能抑制宫颈癌SiHa细胞增殖,并促进其凋亡,其机制可能与Bcl-2/Bax的比值下调有关。Objective:To investigate the effects of different concentrations of Liangxue Huoxue Prescription on proliferation and apoptosis of cervical cancer SiHa cells.Methods:SiHa cells were cultured in vitro,and appropriate cell numbers were selected for plating.Liangxue Huoxue Prescription(concentration 1.125,2.228,3.31,4.37,5.41 mg/ml)was applied to SiHa cells for 24 h,48 h.The proliferation of the cells was detected by MTT and CCK-8 methods.The apoptosis rate of the cells treated with the drug for 24 h was detected by flow cytometry,and the cell state was observed by light microscopy.Finally,the expression of Bcl-2 and Bax were detected by Western-blot.Results:The results of MTT and CCK-8 showed that except for 1.125 mg/ml,the inhibition rate of proliferation was significantly higher than that of the control group(P<0.05),and presented dosage dependence relations.After 48 h of treatment,except for 1.125 mg/ml in MTT,the inhibition rate of each concentration was significantly higher than that of the blank control group(P<0.05).Under light microscopy,except for 1.125 mg/ml,the other groups showed that the volume of the cells became smaller,deformed,wrinkled,rounded and other apoptotic forms.The results of flow cytometry showed that all the concentrations of Liangxue Huoxue Prescription can promote the apoptosis of SiHa cells.Western-blot results showed that the ratio of Bcl-2/Bax was down-regulated except for 1.125 mg/ml.Conclusion:A certain concentration of Liangxue Huoxue Prescription can inhibit the proliferation of SiHa cells and promote its apoptosis.The mechanism may be related to the down-regulation of Bcl-2/Bax.
