摘　　要：目的探究MMP-2、CD133及Ki-67在骨巨细胞瘤组织中的表达情况以及价值分析。方法选取2015年4月至2017年3月本院收治的60例GCT患者,采用免疫组织化学染色检测MMP-2、CD133及Ki-67蛋白在GCT患者中的表达,并且分析这3种蛋白的表达与GCT临床病理特征以及预后的关系。结果 MMP-2阳性产物主要位于肿瘤细胞浆内及胞膜,呈棕黄色、棕褐色颗粒,阳性率为48.33%(29/60)。CD133阳性定位于细胞膜或细胞间质,呈棕褐色,阳性率为43.33%(26/60)。Ki-67阳性定位于细胞核,呈棕黄色,结果显示阳性率为58.33%(35/60)。MMP-2、CD133及Ki-67的表达与肿瘤直径、Campanacci分期以及Jaffe分级有关,差异有统计学意义(P<0.05);GCT患者中术后复发率为18.33%;2年内,MMP-2阳性者无复发生存率为81.00%,阴性无复发生存率为91.00%;CD133阳性者无复发生存率为82.00%,阴性无复发生存率为93.00%;Ki-67阳性者无复发生存率为76.00%,阴性无复发生存率为92.00%,差异有统计学意义(P<0.05)。结论 MMP-2、CD133和Ki-67表达与GCT患者的预后密切相关,对于评估患者的病情以及预后具有一定的参考价值。Objective To investigate the expression and value of MMP-2,CD133 and Ki-67 in giant cell tumor of bone.Methods 60 patients with GCT admitted to our hospital from April 2015 to March 2017 were enrolled.Immunohistochemical staining was used to detect the expression of MMP-2,CD133 and Ki-67 protein in patients with GCT.The relationship between protein expression and clinicopathological features of GCT and prognosis.Results The MMP-2 positive products were mainly located in the cytoplasm and membrane of the tumor,and were brownish-yellow and brown-yellow.The positive rate was 48.33%(29/60).CD133 was positively localized to the cell membrane or interstitial,and was brown,with a positive rate of 43.33%(26/60).Ki-67 was positively located in the nucleus and distributed in brown-yellow particles with a positive rate of 58.33%(35/60).The expressions of MMP-2,CD133 and Ki-67 were correlated with tumor diameter,Campanacci staging and Jaffe grade,and the difference was statistically significant(P<0.05).The postoperative recurrence rate was 18.33% in patients with GCT.Within 2 years,MMP-2 positive patients had a recurrence-free survival rate of 81.00%,a negative recurrence-free survival rate of 91.00%;CD133-positive patients had a recurrence-free survival rate of 82.00%,and a negative recurrence-free survival rate of 93.00%;Ki-67-positive persons The recurrence-free survival rate was 76.00%,and the negative recurrence-free survival rate was 92.00%.The difference was statistically significant(P<0.05).Conclusion The expression of MMP-2,CD133 and Ki-67 is closely related to the prognosis of patients with GCT,and it has certain reference value for evaluating the patient’s condition and prognosis.
