摘  要:目的观察MG-63骨肉瘤细胞在缺氧条件下肾上腺髓质素(ADM)和血管内皮生长因子(VEGF)的表达,并分析两者的相关性。方法将MG-63细胞分为4组:正常培养组,6 h、12 h和24 h缺氧诱导组,缺氧诱导6,12,24 h,分别应用real-time PCR法检测MG-63细胞中ADM和VEGF mRNA表达,Western blotting法检测ADM和VEGF蛋白表达。结果缺氧诱导下MG-63骨肉瘤细胞的ADM和VEGF mRNA和蛋白表达均增强,与正常培养组相比差异有统计学意义(P<0.05)。随着缺氧诱导时间的延长,两者的表达同步增强。结论在缺氧条件下,骨肉瘤细胞的ADM和VEGF的表达被激活,且两者的表达同步增高。Objective To observe the expression of adrenomedullin(ADM)and vascular endothelial growth factor(VEGF)in MG-63 osteosarcoma cells under hypoxia conditions,and analyze the correlation between them.Methods MG-63 cells were divided into four groups:normal control group,and 6,12,24 h hypoxia groups.The expression of ADM and VEGF mRNA and protein in MG-63 cells was detected by real time PCR and Western blotting at 6 h,12 h and 24 h after hypoxia,respectively.Results The expression of ADM and VEGF mRNA and protein in MG-63 osteosarcoma cells was enhanced in hypoxia groups compared with normal control group(P<0.05).With the prolongation of hypoxia induction time,the expression of ADM and VEGF was increased synchronously.Conclusion ADM and VEGF expression in osteosarcoma cells can be activated under hypoxia condition,and their expression is increased simultaneously.
