摘  要:目的比较采用腹部切口与背部切口进行双侧卵巢切除术建立雌性SD大鼠雌激素缺乏模型的优缺点。方法12周龄雌性SD大鼠20只,各10只分别行腹部开口卵巢切除术和背部开口卵巢切除术,比较两种手术方法的切口大小、手术时间、术后恢复时间及术后并发症的情况。涂片检查阴道脱落细胞确定动情周期,ELISA法检测外周血血清雌二醇含量变化,判定大鼠雌激素缺乏模型的建立是否成功。结果背部开口术式的切口长约1.0-1.5 cm,手术时间为1.0 h,术后7-10 d伤口可恢复,无肠梗阻等严重并发症出现。腹部开口术式切口长约1.5-2.0 cm,手术时间为1.5-2.0 h,术后10-14 d伤口可恢复,1只大鼠术后并发肠梗阻。大鼠的动情周期分为动情前期、动情期、动情后期和动情间期。术前各个时期内的阴道脱落细胞形态及外周血雌二醇含量呈动态变化。术后2周阴道脱落细胞涂片检测显示大鼠动情周期变化消失。术后检测血清雌二醇含量较术前明显降低(P<0.05)。结论术后大鼠外周血雌二醇含量及阴道脱落细胞镜检结果提示,两种手术方法都可以成功建立雌激素缺乏大鼠模型。而且雌性SD大鼠背部开口双侧卵巢切除术具有简单易行、创伤较小、可重复性好和成功率高的优势,是进行雌激素相关疾病机制和防治研究的理想模型。Objective To compare the advantages of back-incision ovariectomy versus abdominal-incision ovariectomy for establishing the estrogen deficiency rat model.Methods Twenty 12-week-old female SD rats were randomly divided into back-incision ovariectomy group and abdominal-incision ovariectomy group.The incision size,the operative time,the postoperative recovery time and the postoperative complications were compared between two surgical methods.The smear of vaginal exfoliated cells was observed to determine the physiological cycle,and the serum estrogen of peripheral blood was detected by ELISA.The vaginal exfoliated cells and the content of estradiol before and after surgery were measured to determine whether the estrogen deficiency model is successfully established.Results In the back-incision ovariectomy,the incision size was 1.0-1.5 cm,the operative time was 1.0 h,the recovery time was 7-10 d,and no serious complication such as intestinal obstruction was found.In the abdominal-incision ovariectomy,the incision size was 1.5-2.0 cm,the operative time was 1.5-2.0 h,the recovery time was 10-14 d,and the intestinal obstruction was found in one rat after the surgery.Rat estrous cycle is divided into proestrus,estrous,metestrus and diestrus.The morphology of vaginal exfoliated cells and the serum estrogen content of rats showed dynamic changes during the estrous cycles before surgery.Two weeks after ovariectomy,the vaginal exfoliated cell smears showed no significant change.And the estrogen content after ovariectomy was significantly lower than that before operation(P<0.05).Conclusion According to the postoperative estradiol content in peripheral blood and the vaginal exfoliated cell smears,the estrogen-deficient rat model is successfully established by two methods.By comparison,the back-incision bilateral ovariectomized female SD rat model has the advantages of simple and easy operation,less trauma,nice reproducibility and high success rate,and it can be an ideal model for the mechanism and prevention research of estrog
