摘 要:目的:通过探究转染DCN基因对成纤维细胞TGF-β1、Smad3、Smad7表达及细胞增殖的影响,探讨DCN基因对TGF-β1/Smads信号通路的调控作用机制。方法:采用Real time PCR检测TGF-β1、Smad3和Smad7基因的表达变化。结果:通过对转染后的成纤维细胞进行Real time PCR检测、细胞增殖检测:转染组与对照组相比,DCNmRNA、Smad7mRNA表达量明显增加,而TGF-β1mRNA、Smad3mRNA表达量明显减少,差别均有统计学意义(P<0.05)。结论:通过转染DCN基因对COPD大鼠气道成纤维细胞影响的实验观测(TGF-β1、Smad3和Smad7)基因的表达变化,转染DCN基因对TGF-β1/Smads信号通路产生抑制作用,干预成纤维细胞增殖速度。Objective:To explore the regulatory mechanism of DCN gene expressed on TGF-beta 1,Smad3,Smad7 and cell proliferation in fibroblasts by investigating the effect of transfected DCN gene on TGF-beta 1/Smads signaling pathway.Methods:The real time PCR was used to detected the expression changes of TGF-beta 1,Smad3 and Smad7 genes.Results:The transfected fibroblasts were detected by Real time PCR and cell proliferation:compared with the control group,the expressions of DCNmRNA and Smad7mRNA were significantly increased in the transfection group,while the expressions of TGF-beta 1mRNA and Smad3mRNA were significantly decreased in the transfection group,with statistically significant differences(P<0.05).Conclusion:Through the experimental observation of the influence of transfected DCN gene on airway fibroblasts of COPD rats(TGF-beta 1,Smad3 and Smad7 gene expression changes,the transfected DCN gene inhibited TGF-beta 1/Smads signaling pathway and interfered with the proliferation rate of fibroblasts.
