摘 要:目的:检测人胃腺癌组织中环氧合酶-2(COX-2)、诱导型一氧化氮合酶(iNOS)、核转录因子-κBp65(NF-κBp65)蛋白的表达。方法:采用免疫组化法检测85例胃腺癌组织及其癌旁正常胃黏膜组织中COX-2、iNOS、NF-κBp65蛋白的表达情况。结果:胃腺癌组织中COX-2、iNOS及NF-κBp65蛋白的阳性表达率均高于癌旁正常胃黏膜组织(P<0.001)。胃腺癌组织中COX-2、iNOS、NF-κBp65蛋白阳性表达率与胃腺癌浸润深度和有无淋巴结转移有关,浸润深肌层组阳性表达率高于浸润浅肌层组,有淋巴结转移组高于无淋巴结转移组,且COX-2蛋白在高、中分化组中的阳性表达率高于低分化组(P<0.05)。胃腺癌组织中COX-2、iNOS与NF-κBp65蛋白的表达呈正相关(r P=0.366、0.359、0.405,P<0.001)。结论:胃腺癌组织中COX-2、iNOS、NF-κBp65蛋白表达升高,三者参与胃腺癌的形成且与其侵袭、转移有关。Aim:To detect the protein expressions of cyclooxygenase-2(COX-2),inducible nitric oxide synthase(iNOS)and nuclear factor-κBp65(NF-κBp65)in human gastric adenocarcinoma tissue.Methods:Expressions of COX-2,iNOS and NF-κBp65 protein were detected by immunohistochemistry in 85 gastric adenocarcinoma tissue specimens and adjacent normal gastric tissue specimens.Results:The expressions of COX-2,iNOS and NF-κBp65 protein in gastric adenocarinoma tissue were significantly higher than those in adjacent normal gastric tissue(P<0.001).The positive expression rate of COX-2,iNOS and NF-κBp65 protein in gastric adenocarinoma tissue was related with soaking depth and lymph node metastasis,the positive expression rate of COX-2,iNOS and NF-κBp65 protein in the deep infiltrating muscle group or the group with lymph node metastasis was higher,and the positive expression rate of COX-2 protein in the high and middle differentiation group was significantly higher than that in the low differentiation group(P<0.05).There was a positive correlation between the expressions of COX-2,iNOS and NF-κBp65 in gastric adenocarcinoma tissues(r P=0.366,0.359,0.405,P<0.001).Conclusion:The protein expressions of COX-2,iNOS and NF-κBp65 in gastric adenocarcinoma tissue are high,which might promote carcinogenesis,invasion and metastasis.
